In this study the dormancy release reasons of Abies holophylla Maxim are discussed, based on the test of morphological characteristics, seeds of seed capsule of permeability, kind of seed capsule and endosperm of distilling variations and inhibit determination, as Abies holophylla Maxim the research object. While using different layer deposition temperature and different GA3 concentration of seed handling and their interaction processing, exploring the best method to remove the dormancy of Abies holophylla Maxim. In the process of stratification, measuring the activity of soluble sugar and starch, soluble protein, amylase and G6PDH and physiological and biochemical indexes such as the content of endogenous hormones, through taking specimen regularly, and then understanding the mechanism dormancy and germination of Abies holophylla Maxim. The results were as follows:1,The seed capsule of Abies holophylla Maxim has poor permeability, and the maximum permeable rate is 4.868%,the seed has mechanical side effect on the seed germination.2,Through extraction determination the kind of seed capsule and endosperm variations and inhibit substances of Abies holophylla Maxim, the kind of seed capsule and endosperm all contains germination inhibitors substances, they had significant inhibitory effect on the germination rate, root seedling height and growth of the cabbage seed, and the higher the concentration, the more significant inhibitory effect. The endosperm inhibitory effect is bigger than the kind of seed capsule.3,In the process of stratification of Abies holophylla Maxim seed, soluble sugar,starch and soluble protein are in continuous transformation and utilization status, their content are all in an down trend after the first 10 days of stratification.At the 10 to 30 days, their contents have the upward trend.Infer at the first 10 days of stratification, the seeds removed dormancy, from the 10 days the seeds turn to the germination conditions.4,At the stratification, the activity of a-Amylase has the upward trend, but its continuous process is fluctuated.The activity ofα+β-Amylase has the upward trend, its maximum ascensional range appears at the first 10 days. The trend of G6PDH activity turn down at the first and then upward, and its reach the peak value at the 30 days, at this time its G6PDH activity higher then the initial period of the stratification.5,The content of endogenous IAA and GA3 has a tendency of rising with time increasing but fluctuated in the process.The content of GA3 has the maximum ascensional range at the first 10 days of the stratification.And the content of ABA has fall down obviously during the first 10 days of the stratification.At the same time, the ratio of GA3/ABA and IAA/ABA generally has a tendency of rising at the first 10 days of the stratification.
