The experiment was conducted to investigate the effect of betaine and biotin on fatty liver hemorrhagic syndrome(FLHS) in laying hens. Two hundred and twenty five Hy-line brown laying hens at the age of 300 days were randomly assigned to 3 groups, with 3 replicate pens per group and 15 laying hens per pen, and a feeding experiment was conducted with them for 60 days. The laying hens were fed with either normal diet(ME 11.30MJ/kg and CP 17.20%), high energy-low protein diet(ME 12.81 MJ/kg and CP 13.04%), the high energy-low protein diet plus betaine, the high energy-low protein diet plus 0.3mg/kg biotin, the high energy-low protein diet plus betaine and biotin. Feed and water were provided for ad libitum consumption and the birds received light of 16h a day. Six hens from each group were selected on 1 st,30th, and 60th days to collect samples of muscle, respectively. The concentration of rate of muscle fat, laying performance and histopathologic changes were investigated to determine the effect of betaine and biotin against FLHS, and the relative expression levels of muscle apoA I and apoB100 mRNA were investigated by real-time fluorescent quantitative PCR to reveal its potential mechanisms. The results showed that compared with the control group, feed the laying hens with high energy-low protein diet for 30 days could induce FLHS, and feeding for 60 days could lead to severe FLHS. Feed the laying hens with high energy-low protein diet plus betaine and biotin significantly prevent the inducion of FLHS. Compared with the control group, feed the laying hens with high energy-low protein diet for 30 days significantly decreased the expression levels of muscle apoAâ… mRNA and muscle apoB100 mRNA, and feeding for 60 days significantly decreased both the levels of muscle apoAâ… and apoBioo mRNA. Both the relative expression levels of muscle apoAâ… and apoB100 mRNA were significantly increased by the addition of betaine and biotin into the high energy-low protein diet.
