The polysaccharides from Coprinus comatus were isolated by ultrasonic or hot water, and its chemical structures were analysised by HPLC, GC, GC/MS, AFM and so on. The effect mechanism of ultrasound on the structure and biological activity of polysaccharide islated from Coprinus comatus and its structure-activity relationship were studied.(1) Study on the Feasibility of ultrasonic effect to extract the Polysaccharide from Coprinus comatus in the viewpoint of the antioxidant activity in vitro. The Corpinus comatus polysaccharides are extracted by hot water and ultrasound. Different polysaccharides have some functions of scavenging free radicals and inhibiting lipid peroxidation. The Corpinus comatus polysaccharides extracted by ultrasonic-assisted have a better antioxidant activity. The results indicated that the ultrasonic radiation has feasibility in the extraction of plant polysaccharides.(2) The chemical structures of WCP3a and UCP3a were analysised by HPLC, GC, GC/MS and their antioxidant activity in vitro were also measured. The results indicated that (1) The WCP3a was composed of mannose, glucose and galactose, but UCP3a was composed of mannose and glucose; (2) WCP3a contained (1→4,6)-linked-α-D-mannitol, (1→)-linked-α-D- mannitol, (1→>3)- linked-α-D-glucitol, and (1→2,6)-linked-α-D-galactosyl residues, Comparing with UCP3a which containing (1→4,6)-linked-α-D-mannitol, (1→)-linked-α-D-mannitol, (1→3)-linked-α-D-glucitol, and (1→4)-linked-α-D-glucitol residues.(3) WCP3a and UCP3a can remove the Superoxide anion effectively, and the antioxidant activity of UCP3a in vitro was higher than that of WCP3a; Galactose, the monosaccharide composition of polysaccharide isolated from Coprinus comatus, was transformed to glucose treated by the ultrasound.(4) The single-helix structure of Coprinus comatus polysaccharide was determined by combining Congo red test and atomic force microscopy experiment; Polysaccharides isolated from Coprinus comatu had an effect on superoxide anion radical scavenging in the experiment range using pyrogallol self-oxidation; The antioxidant activity of WCP3a isolated from Coprinus comatus in vitro is more closely with its spatial structure.
