Coprinus comatus is a kind of edible and medicinal fungi and the polysaccharide is one of the main active constituents. Recent advances demonstrate that polysaccharides extracted from Coprinus comatus exhibited such activities as enhancement of immune system, antitumor, hypoglycemic, hypolipidemic and antioxidant. At present, more research were focused on the extraction technology, structure and biological activity of its polysaccharides, but there are some problems with the products such as lower yield, deep color and easy absorption of moisture. In this study, some enzymes were added to improve the yield of polysaccharide. Then the enzyme extraction parameters were optimized. In order to improve the products character, the ethanol precipitation process was optimized.the conclusion has the important practical significance and application value for the development of the product.I study the isolation,structural characterization and bioactivity of glucan from Coprinus Comatus, it lay a foundation for the research of fungi polysaccharide depth structure-activity relationship.1.Enzymatic extraction and ethanol precipitation technologyI find that papain was the best enzyme and its optimal conditions were as follow:the enzyme reaction temperature50℃, the reaction time7h,enzyme proportion1:100(papain:raw materials). The conclusion of ethanol precipitation technology showed that: concentration methods of boiling, concentrate ratio of1:2, washing frequency of1time, dry way of vacuum freeze-drying.the polysaccharide yields reach to2.96%, raising45.1%compare with the traditional method which yields reach to2.04%. In addition,both could stimulate proliferation of macrophage, so this method could be applied in Large-scale industrial production.2.Study on structure elucidation immunological activity of high molecular weight polysaccharidesfruiting body of Coprinus comatus were crushed to powder, followed by extraction with hot water, concentrated with ultrafiltration,40%ethyl alcohol sank the polysaccharide fractions CCP750K-40P, In addition, it could stimulate proliferation of macrophage.The structures of CCP750K-40P were investigated by composition analysis, methylation analysis, IR, NMR spectroscopic methods (1D NMR:1H NMR,13C NMR, 2D NMR:COSY, TOCSY, HSQC, HMBC and NOESY spectra), it with a molecular mass of264×104Da, contained glucose.
