Astragali Radix, the dry root of Astragalus mongholicus Bge, has been used for centuries as an important medicine to reinforce vital energy, and to promote the discharge of pus and the growth of new tissue. Since Astragalus mongholicus Bge,has a long cultivation cycle, are greatly impacted by the environment production can be unstable and growers can hardly satisfy the market demand. The establishment of suspension cell lines is the best solution to address this problem. This study uses MS medium and adjusts the ratio and concentrations of hormone to establish the cell suspension line. Methyl jasmonate has been supplemented to improve the content of flavonoidsThe results are as follows:1.The best Astragalus mongholicus Bge. explants for cell suspension line is the hypocotyl.2.Cell suspension line of Astragalus mongholicus Bge. has been established for the first time, the best culture condition is:MS +1.5 mg·L-12,4-D +2.0 mg·L-1 6-BA +30% sucrose; pH 5.5~6.0; initial inoculum 50 g·L-1 and generation inoculation 37 g·L-1.3.Studied the characteristics of suspension cells Astragalus mongholicus Bge.. The cell growth curve has been drawn and growth cycle is 25 days.Lag phase is 1 to 10, the logarithmic growth phase is 11 to 19,the 19 d make the cell biomass reached maximum, resting stage is 20 to 22 and the decline phase is 23 to 25. pH value of medium began to rapidly dropped to 4.5 and back to 6 or so. Then it in the relative stable value around 5.7. The cell activity in the logarithmic phase and lag phase is stronger than in the early lag phase and decline phase.It shows that the optimal period of flavonoids substances is late lag phase and the logarithmic growth phase.4. Methyl jasmonate has a significant effect on growth of cell and flavonoid content of Astragalus mongholicus Bge..The result showed that the best concentration is 10μmol·L-1,make the dry weight of the cell up to 20.76g·L-1, flavonoid content of cell is up to 3.41 mg·g-1,flavonoid content of medium reached 1.15 mg·g-1.This concentration is better than control and others.The best time for MeJA is the 7th day, in this period,the dry weight of cells reached 21.16 g·L-1,flavonoid content of cells is 3.25 mg·g-1,flavonoid of content medium was 1.31 mg·g-1,this provided the basis for commercial production.
