This study was initiated to induce callus from radicle,hypocotyl,cotyledon which formed the seeds of Astragalus mongholicus Bge.and the acquisition of the young embryo explants,It explored regulatory factors for Astragalus mongholicus Bge.callus growth and flavonoids synthesis from the following aspects:the medium composition,culture conditions,organic appendages,different elicitors etc,and aimed at improving Astragalus mongholicus Bge.callus biomass and the content of flavonoids in the laboratory conditions,the selected high-yield Flavonoids in cell lines for the industrial production of Astragalus mongholicus Bge.to provide the oretical support and technical guidance.The main findings are as follows:1.Astragalus mongholicus Bge.types of explants on callus induction has a significant impact,this three kinds of Radicle,hypocotyl,cotyledon explants can induce callus,The rate of induced callus of cotyledons and hypocotyls was significantly higher than radicles,the lower the rate of radicles induction,the callus browned serious,easy to aging and growed very slowly.2.Selected hypocotyls as explants and MS as the basic medium,Effects of 2,4-D,NAA,6-BA on callus induction were investigated by using the single and combined use of two methods to analysis and comparison of the various factors in callus induction in the same material between different concentrations,selected callus induction best growth regulators combination of Astragalus mongholicus Bge.for the MS + 1.0 mg/L/6-BA + 2.0 mg/L2,4 -D.3.Growed about 30 days with and without suspensor in immature embryos of Astragalus mongholicus Bge.were inoculated with three different medium,B5 medium selected is one of Astragalus mongholicus Bge.'s immature embryo optimum callus induction medium;At the same time,the experiment confirmed the existence of Suspensor for survival of Astragalus mongholicus Bge.'s immature embryo is not a key factor.4.The adoption of single-factor test culture conditions studied,sugar concentration,nitrogen source,organic appendages hydrolysis of casein protein, elicitor methyl jasmonate for Astragalus mongholicus Bge.callus growth and flavonoids accumulation,determined Astragalus mongholicus Bge.callus growth and accumulation of flavonoids in the optimum conditions for subculture to MS + 6-BA2.0mg / L +2,4-D1.0 mg / L + NAA1.0mg / L + sucrose 4%for the medium, pH in the 5.6~5.8 between,12h / d light;NH4+:NO3- as the best nitrogen source for 1:1;determined the hydrolysis of casein concentration of the best add 1g / L; jasmonate concentration of the best add 1μmol / L.
