As one of the precious farmed fur animals, mink's main economic value was its fur. This study analyzed the difference expression of fibroblast growth factor-5 (FGF5) gene, at the level of mRNA, in difference of minks' breeds; and FGF5 distribution in different body tissues of mink. The results revealed that molecular mechanism of FGF5's impaction on coat length.The main contents of this experiment were as follows:1. Measurement and analysis of various mink coat of length, two breeds- Dalian Jinzhou black mink and American black mink were selected with 60 minks each, half male, half female. Measured and data analyzed the difference varieties of each mink of hair's length.The results showed that, in mid-back, neck cross and both sides of belly; the hair's length of male minks was longer than the same kind of females. Compared the guard hair's length of same sex of mink, Jinzhou black minks were longer than American black minks. At the both sides of belly fluff's length of male American black mink was longer than other groups; the fluff's length of Jinzhou black mink was longer than female American black mink; this results all showed significant difference (P<0.01). But the result showed no significant difference (P>0.05) of other contrasts.2. The study analyzed the difference of FGF5 gene expression at the level of mRNA , According to sex and breeds, the experiment divided mink into 4 groups (male Jinzhou black minks, female Jinzhou black minks, male American black minks and female American black minks), and randomly chosen 3 biological individuals, The test was analyzed FGF5 gene expression on the level of mRNA, which using FQ-PCR, on mink.The results showed that, FGF5 gene expressed mRNA, was both in brain tissues and skin tissues. The expression in brain tissue was significantly higher than in skin tissue, in the same sex and breed, (P<0.01); FGF5 gene expression on the level of mRNA, was negative correlation in skin and brain. In the same sex of mink, the expression of FGF5 gene on the level of mRNA in American black minks was higher than it in Jinzhou black minks.3. The study analyzed FGF5's qualitative and localization in mink's body tissues; Choosing brain tissues and skin tissues, paraffin and frozen section methods were used to observe hair follicles structures and its development. By using Rabbit Anti-FGF5 as monoclonal first antibody, Goat anti-Rabbit, HRP conjugated as second antibody, of frozen-section, experiment did immunohistochemi- stry by the [SP] method, and also did indirect immunofluorescence by using Cy3 and FITC.The results showed that, under light or fluorescence microscopy, FGF5 was expressed in both brain tissues and skin tissues of mink, in mink's same tissue the content of FGF5 in brain was higher than it in skin. The content of FGF5 was the same tendency with FGF5 gene expression on the level of mRNA.Conclusion: Mink of hair's length were a reverse trend with FGF5 gene expression on the level of mRNA. In same sex and breed, FGF5 gene expression on the level of mRNA in skins was negative correlation with it in brain; the content of FGF5 were the same tendency with FGF5 gene expression on the level of mRNA, that showing FGF5 gene at the post-transcription level had no larger variety.
