Study On Affect That Sprout Brings To Flavonoids Synthesis And Anti-bacterial And Anti-tumor Ctivity On Germinated Tartary Buckwheat

This subject took tartary buckwheat seeds as materials, in order to observe the effects of every condition such as ultrasonic, changing the conditions of soaking, increasing ultraviolet irradiation during tartary buckwheat germination process and thus to determinate total flavonoids content and related enzyme with respective study of the correlation of flavonoids synthesis amount and different germination factors and the enzymes activity in synthesis methods. Besides, we also investigated the effect of tartary buckwheat germination extract (TBGE) on anti-bacterial, anti-allergic and anti-tumor activity.1,On basis of this laboratory research , we germinated tartary buckwheat by using ultrasonic, soaking with CuSO4 and ZnSO4, and increasing ultraviolet irradiation with the pertinent temperature and humidity. After 7days, the results showed that ultrasonic for 6 minutes soaked with 0.01%CuSO4 and 0.05% ZnSO4 and could make seeds have the highest germination rate, germination vigor, and the fresh weight of tartary buckwheat sprout. Ultraviolet irradiation had no appreciable impact on germination rate, but ultraviolet irradiation 1 hour every day could make a higher fresh weight.2,Ultrasonicing for 6 minutes, soaking with 0.01%CuSO4 and 0.05% ZnSO4 and ultraviolet irradiation 1 hour every day could obviously stimulate the synthesis of secondary metabolites, at the same time improving flavonoids cargoes. Total flavonoids content of tartary buckwheat germination extract (TBGE) have an upward trend along with the germination time. During the whole germination, the change of Phenylalanine ammonia-lyase (PAL) activity is quadratic curve but that of Chaleone isomerase (CHI) had a linear relation; it had no correlation between that. Rapidly increases of Buckwheat flavonoids content is the result of a combined action of PAL and CHI in germination prophase. Along with the germination lengthening, the elevation of CHI activity is the major cause for the increase of flavonoids content.3,The main components of flavonoids are rutin and quercein and we choose HPLC method to detect their content. And with the extension of germination (1~7d), the content of rutin was raised while the content of quercein was reduced. One thing to be mentioned is that on the third day, the proportion of rutin and quercetin was close to 1:1.4,Different TBGEs had different inhibitory effect on the test fungi. TBGEs had a higher inhibition effect on Strawberry Botrytis cinerea than Alternaria Nees ex Wallr, but no effect on yeast and Penicillium glaucum. The inhibition ratio had dosage effect with content of flavonoids. TBGEs had inhibition effect on the formation of conidia and also they reduced content of soluble protein in mycelium.5,This thesis investigated the effect of TBGE on anti-allergic reaction of the rodent experimental models. The extracted Wistar rat peritoneal mast cells (RPMC) in vitro was used to test the effects of TBGE with different germinating periods on histamine releasing. Results showed that the TBGEs could inhibit the release of histamine from RPMC responded to Compound 48/80. This effect of inhibition of TBGEs was much higher than the extracts from seeds, and among these TBGEs, the 3d greminated sprouts had the highest inhibition effect. Rutin and quercetin both manifested the antihistamine effect. And compared with rutin, quercetin had a higher synergistic effect.6,The anti-tumor action of TBGEs on human lung cancer cell line A549 was examined, using a modified MTT assay. The tested samples exhibited significant inhibitory effect on A549 cell proliferation; particularly, the TBGE from germinated 3 days had the highest inhibition effect. Rutin and quercetin both had inhibitory effect. And compared with rutin, quercetin had a higher inhibition ratio.
