| More and more environment damages were displaied by increasing usage of chemical synthesis pollution and plants'. People are paying more attention to the needs of natural products. The active ingredients of natural plant have become one of the major research objections. Some report that Halianths Tuber leaves contain natural flavonoids, which is a sort of important resource for the development of natural antioxidants. The optimum extraction conditions of flavonoids were studied by single factor and orthogonal experiment with these Halianths Tuber leaves. Then the physiological activities of Halianths Tuber leaves flavonoids were conducted, including stabilities, antioxidant activities, anti-tumor activities and anti-bacterial activities et al.The main contents and results are as follows:1. The study of optimum extraction conditions:The optimum conditions of normal reflux extraction was that: ethanol concentration 80 % , material to ethanol 1:25, extracting temperature 90℃and extraction time 2h.The extracting rate is 1.411%.The optimum condition of ultrasonic extraction was that:material to ethanol 1:20, ethanol concentration 60%, extraction time 30 min and extracting temperature 80℃,The extracting rate is 1.921%.The optimum condition of microwave assistant extraction was that:ethanol concentration 50%,extraction time 5min, material to ethanol 1:30 and power 300w:The extracting rate is 2.481%.By compare three extracting methods, it is found that microwave assistant extraction method is more effective than that of the other extracting methods.2. The studies of biological activities:Study of stability activities: Halianths Tuber leaves flavonoids have good stability against heating. However, it should be stored under the environment without strong light, oxidizer, deoxidizer and alkaline substances.Study of antixoidative activities:Halianths Tuber leaves flavonoids show good activities. It had exhibited good resistance to lipid peroxidation.Study of antitumor activities: The growth of KB, HepG-2, K562 tumor cells is inhibited by Halianths Tuber leaves flavonoids significantly in dose-dependent manner.Study of antibacterial activities: By combining inhibition zone and MIC , The results are as follows: Staphylococcus aureus>E.coli>Bacillus subtilis... |
