Poplar canker is a destructive disease which is hard to control. This experiment was done through tissue culture and physiological and biochemical technology method, cultivating callus tissue as experimental material,and they were handled four times by toin ,finally, the callus tissues got the resistance,analysis them from physiological and biochemical test: the activity of peroxidase, superoxide dismutase, phenylalanine ammonia-lyase, polyphenol oxidase, chitinase, with a view to reveal the toxin resistance of the most important factors.(1) In this research,when the callus tissue were handled by toxin,the activity of enzyme changed a lot afer the handle,and the trend of the activity are rise first and then desend. In the callus tissue which were handled by toxin,the activity of the phenylalanine ammonia-lyase is rising fast,and then drop with the time flys,the change is big,its biggest activity is five times than before; The activity of peroxidase got the apex when the callus tissue were handled threr times,and the range were big; The activity of superoxide dismutase,in its third time,the activity was(1964.914) ; and the contrast was(1203.693); From the first generation to fourth,the activity of polyphenol oxidase was always higher than contrast,and the summit appeared in the third time; The activity of the chitinase has distinct changes,and it got the summit in the second time,the rate is one hundred and thirty five percent,ang in the fourth time, it's sixty two percent.From the result,we could find that the callus tissue was handled by toxin,the activity of enzyme rise,and with the time flying,the general activity is higher than the contrast,we could find,the callus tissue has got the resistance to avoid the toxin invading,and it helps us to find the new resistant type of poplar.(2)Through testing we could found,though the enzyme activity were different after treated,but in these four periods,but they occurred distinctly changes. SOD,PPO and POD achieved maxium in the third time;these emyme had the same pattern,rose first and then dropped;but they aren't synchronize,this result may have the relationship with inner difference.(3)From the results,we found when the callus tissue were invaded by canker, the activity of PAL,PPO and POD rose,SOD and Chitinase composed,improved the strength of cell wall,finally,induced the resistance of canker.
