In order to eluciate the mechnism of callus differnciation and understand the mechnism of organogenesis, physiological and biochemical change characteristics during the course of plant regeneration in rice tissue culture were studied with 253(restoring line) and BoB(male sterile lines) used widespreadly as research materials. The results showed as follows.1. Callus inducation percentage of 253 and BoB was 85.7% and 90%, respectively, there was not significant difference between two rice materials. Desiccation improved differentiation rate of callus, 2 d desiccation treatment had best effect, and callus differentiation rate of BoB and 253 was 87% and 92%, respectively.2. O2- content of BoB and 253 decreased from 1st day to 5th day during callus differentiation process, it were 85.9 μmol·g-1 FW and 124.3 μmol·g-1 FW, that of 253 was significantly higher than that of BoB, and there was not significant difference after 5th day.
