Polymorphism of 23 tree peony cultivars was studied by the technique of AFLP(Amplified Fragment Length Polymorphism). The improved method of SDS-DNA isolation was used to extract total genic DNA from old leaves of Paeonia suffruticosa, High quality total genic DNA was obtained. The values of A260/A280 of each sample were berween 1.7 and 1.8. Strips in agarose electrophoresis were clear and less decomposed. These DNA samples with high purification were intact without containing inhibitor of enzyme reaction and could be completely digested by restriction enzymes such as Mse â…  and Pst â… . The system suitable for silver-stained AFLP analysis were established successfully by restriction enzymes digesting-ligasing reaction, preamplification,selective amplification. Sharp fingerprintings were obtained by thses processes.243 loci bands in genome DNA were detected by 8 primer combinations and the average polymorphism ration was 80.3%. The dendrogram generated result by SPSS11.5 software Average Linkage(within group)showed: these 23 cultivars were divided into 2 culsters. One was composed of cultivars from Group of Central Plains of China and another was composed of cultivars from Group of Japanese; cultivars of the same species origin were divided into the same group,which shows the origin of cultivars was more important than other characters on the relationship of tree peony cultivars,which was constent with the conclusion of Mengli. At the same time, the result of proved the conclusion from the molecule level that the species origin is the precondition standard of cultivars classification.The dendrogram showed that these 2 clusters were not consistent with the classification result generated by the flower type, colour and other characters of tree peony configuration.The cultivars of the same flower type didn't compeletely exit in the same cluster and the cultivars of the same flower colour didn't exit in the same cluster,either. These cultivars were classified into the same cluster only when other configuration characters were the same or similarity. From this we can conclude that AFLP is more genuine on the relationship of tree peony cultivars than the single character of configuration,so,AFLP is one of the suitable techniques for classification of tree peony cultivars.
