| Paulownia is one of the fast-growing trees. It is important to plant Paulownia because it not only can solve the problem of the shortage of wood in China but also can improve the ecological environment in global. However, because the paulownia witches" broom which is a disease of trees caused by Phytoplasmas has greatly effect on Paulownia timber productivity, it is difficult to plant it in larger areas. Solving this problem is an important tissue and people have done lots of work all along. But, Phytoplasmas is a small microbe which has no cell wall and it is not easy to be cultured in vitro. People has not yet found a kind of medicine that could kill Phytoplasmas, therefore, the problem of paulownia witche's broom has not been solved so far.This paper dealt the paulownia tissue culture with different kinds and different concentrations of extraneous substances such as FU, Rifampicin, Acheomycin and vitamin C, and we tried to find the suitable combination that could kill Phytoplasmas, and to discuss the mechanism of this disease further more. Through these experiments we found that FU and vitamin C had less effects on paulownia witches' broom , but, the higher concentration of Rifampicin could restrain the growth of Phytoplasmas effectively. But using Rifampicin it only, plants also could be poisoned seriously, and even caused them to death. Using Acheomycin only, it could release the disease at some extents in some days but it could not be culed ultimately. However, when NAA(0.2 mg. L-1 ), Rifampicin (80—120 mg. L-1 ) , Acheomycin50～150 mg. L～(-1)) ,Vitamin C (0～1g. L-1 ) were added to the culture medium together, the diseased paulownia could be culed ultimately and the disease would not recrudence, it had no any toxion action to the growth of plants either. It was improved by PCR that there were no Phytoplasmas any more in the culed plants.But ,by stastical analysis, we got a best combination which was 0.2 mg. L-1NAA+100mg. L-1 Rifampicin+100 mg. L-1 Acheomycin+0 g. L-1 Vitamin C.Throngh the protein 2-SDS-PAGE, we found that protein of pI 6.8, WM 24KD and protein of pI 7.4, WM 15KD existited in the healthy seedings but did not existed in the diseased seedings, and we also foung that whether paulownia witche's broom occur and recudense or not it had relationship with the protein of pI 6.8, WM 24KD. But as for the function of this protein , we have to do some researches further to prove it.
