There are two experiments in this study :one is for morphologic diagnosis method of Eperythrozoon suis(E.suis) , the other is for PCR diagnosis method of E.suis.Experiment 1 The study on morphologic diagnosis method of E.suisThis experiment involves in the study of Wright's,Giemsa and Arcridine methods of blood smear and conformation of E.suis under electron microscope.E.suis compressed tablet of physiological saline under optical microscope are annular, global, cosh, dumbbell and asteroidal shining little bodies, they adhere to the surface of blood cell, singly or conglobately by the shape of chain or squama. There are more than 20 E.suis adhering to every blood cell in severely infected case, at this time the blood cell are by the shape of castor-oil plant and pineapple under optical microscope. The E.suis are dissociateid in blood plasma, they always swing, turn, roll, move up and down, they stop moving once adhering to the blood cell. The less moving bodies look more active.Under optical microscope after Wright's we can see the blood cells are bigger, blood cells and E.suis are dark blue and purple, E.suis are darker, they lies on the extrotrusible position or on the surface of blood cell, 3-8 E.suis on every blood cell.Under optical microscope after Giemsa we can see blue and purple granules of 0.5-1.5μm diameter by roundness and comma , there are many granules on every blood cell.Under fluorescent microscope after Arcridine we can see some bright little bodies on surface and edge of most blood cells of 1μm diameter, which are E.suis dyed by Arcridine.When putting the managed blood under the electron microscope,...
