Due to the importance of insulin-like growth factor-I in regulating many physiology and metabolic processes in animal body, so the IGF-I gene was chosen as a candidate gene to study important economic trait associated polymorphisms in chickens that made it more importance to scientific research. But now, many researches the polymorphisms of IGF-I gene in chickens applied for the PCR-RFLP technology. This technology would be failure in detected the mutation Loci, because some DNA sequence could not be identify by restriction enzyme. PCR-SSCP technology could simple and convenient and fast detected the mutation Loci of gene. So it widely applied. In this paper, we applied PCR-SSCP technology to detect the gene mutation of IGF-I gene in New Yangzhou chickens(130), Xueshan chickens(100) and White Leghorns chickens(47) and DNA sequenced. We discussed the best PCR reaction condition and detection condition of SSCP to assumed gene mutation Loci. The relationship of DNA mutation Loci and traits measured in this experiment were analyzed with GLM model. The results of this experiment as follows.1. In this experiment, we affirmed the best experiment reaction and detection condition in 5'-UTR,exon 1 and 3'-UTR of IGF-I gene and found four mutation Loci (23F/R,26 F/R,28 F/R,29 F/R) in New Yangzhou chickens, Xueshan chickens, White Leghorns chickens, respectively.2. Gene and genotype frequencies were calculated for the polymorphic Loci in IGF-I gene.
