| The polymorphisms of chicken Myf5/Myf6 gene were detected by PCR-SSCP method with two different pairs of primers among 233 chickens from 8 populations (including 5 pure lines and 3 crossbreds developed by Sichuan Animal Science Academy and Dahen Poultry Breeding Company).Based on the detection of polymorphisms,the effects of different genotypes on slauter and meat quality traits were analyzed.There are two SNP sites in the first exon region of Myf5 gene,they are G106T and G182C.This two SNPs were observed in the same primer.Through the haplotype analyzed,the frequence of H1(AC) is 31.85%,the frequence of H2(AD) is 16.00%,the frequence of H3(BC) is 39.39%,the frequence of H4(BD) is 12.75%. The haplotype H1 and H3 have the advantage.The frequence of H1 and H3 are higher than the haplotype H2 and H4.There is a SNP(G47A) in the first exon region of Myf6 gene.The distribution of Myf6 genotype strayed off the Hardy-Weinberg equilibrium and the heterozygosity was low in five purelines but not in three crossbreds.The results by LSM(least square means) analysis and significant test showed that the Live weight(LW),Carcass weight(CW),Brest muscle weight(BMW) and the Leg muscle weight(LMW) of H1H1,H1H3,H3H3 had significant difference with H2H2(P<0.05),and had very markedly difference with H4H4(P<0.01).Each genotype of the Myf6 had significant difference with the Live weight(LW),Carcass weight(CW),Brest muscle weight(BMW) and the Leg muscle weight(LMW)(P<0.05).The wild-type AA had significant positive effects on live-weight, carcass-weight,breast-muscle-weight and leg-muscle-weight(P<0.05).The Fiber density(FD) of H1H1,H1H3,H2H4 and H3H3 had significant lower difference than the H2H2 and H4H4(P<0.05).But the Fiber diameter(FDM) of H1H3 and H3H3 had significant higher difference than the H2H2 and H4H4(P< 0.05).Each genotype of the Myf6 had significant difference with the Fiber density (FD) and the Fiber diameter(FDM)(P<0.05).The Fiber density(FD) of genotype AA and AB had significant lower difference than BB(P<0.05).But the Fiber diameter(FDM) of genotype AA and AB had significant higher difference than BB (P<0.05).The Myf6 had addictive effects and dominant effects of increasing -29.99 and -21.18 of the Fiber density(FD),the addictive effects and dominant effects of increasing 1.025 and 0.835 of the Fiber diameter(FDM). |
