The research adopted the molecular quantitative Genetics metllod , used PCR·SSCP and PCR-RFLPs method to detected 6 exons of 849 GUSHI-ANKA chicken. Gene Sequence and Analysis of the Intron 1,exon 2,exon 4 .Studies on Polymorphisms of Delta-9 Desaturase Gene and their associations with growth traits,slaughter traits,fleshy Traits,muscle traits,blood biochemical index,fat traits,fatty acid contents.Information of major Gene, Gene number, additive effect, molecular marker was developed and evaluated based on data of phenotype and molecular identification of fatty acid trait. The results were as follows.Three SNPs including of Delta-9 Desaturase Gene were detected in F2 Generation.A c.C54T is detected in Intron 1.A c.A495G is detected in exon 2.A c.Gais detected in exon4. They were synonymous mutation.Intron 1 occurred CC,CT,TT genotypes, Exon 2 occurred AA,AG,GG genotypes, Exon 4 occurred GG,GA,AA genotypes. Population Genetics showed that the polymorphic information content of three mutation sites were moderate (0.365-0.375).SNPs genotypes of Delta-9 Desaturase Gene significantly effected the some growth traits. SNPs genotypes of Intron 1 significantly effected the shank girth at the age of 8 and 4 weeks and the breast angle at the age of 12 weeks. SNPs genotypes of exon 4 significantly effected pelvis breadth at the age of 8 weeks,length of breastbone at the age of 8 weeks,the breast angle at the age of 8 weeks.The For Intron1,4, genotypes TT,AA have a positive correlation with these traits.The results of this study suggest Intron1,exon4 of Delta-9 Desaturase Gene can be linkage to controlled growth traits and worthy of further studing as a molecular marker.SNPs genotypes of Delta-9 Desaturase Gene significantly effected the some slaughter traits. SNPs genotypes of Intron 1 significantly effected Claw rate,Gizzard rate,SNPs genotypes of exon 2 significantly effected live weight,Carcass weight,Carcass rate,Semi-evisceration rate,Evisceration weight,Head weight,Evisceration rate,Gizzard rate,Gizzard weight. SNPs genotypes of exon 2 significantly effected Semi-evisceration rate,Breast muscle weight,Carcass rate,Evisceration rate,Pancreas weight.The For Intron1,4, genotypes TT,GG,AA have a positive correlation with these traits. The results of this study suggest Intron1,4 of Delta-9 Desaturase Gene can be linkage to controlled slaughter traits and worthy of further studing as a molecular marker.SNPs genotypes of Delta-9 Desaturase Gene significantly effected the some muscle traits. SNPs genotypes of Intron 1 significantly effected leg muscle weigth,leg muscle girth. For Intron1,genotypes TT have a positive correlation with these traits. The results of this study suggest Intron1 of Delta-9 Desaturase Gene can be linkage to controlled muscle traits and worthy of further studing as a molecular marker.SNPs genotypes of Delta-9 Desaturase Gene significantly effected the some fleshy traits. SNPs genotypes of Intron 1 significantly effected Intramuscular fat band width,sebum,sebum rate. SNPs genotypes of exon 2 significantly effected Breast muscle water loss rate,Intramuscular fat band width,sebum,sebum rate. SNPs genotypes of exon 4 significantly effected leg muscle pH. The For Intron1,2,4, genotypes TT,GG,AA have a positive correlation with these traits. The results of this study suggest Intron1,2,4 of Delta-9 Desaturase Gene can be linkage to controlled fleshy traits and worthy of further studing as a molecular marker.SNPs genotypes of Delta-9 Desaturase Gene significantly effected the some blood biochemical index traits. SNPs genotypes of exon 2 significantly effected low density lipoprotein. SNPs genotypes of exon 2 significantly effected Creatine kinase,Lactic dehydrogenase.SNPs genotypes of exon 2 significantly effected Alkaline Phosphatase,Cholinesterase,Globulin,Lactic dehydrogenase.The For Intron1,4, genotypes TT,AA have a positive correlation with these traits. The results of this study suggest Delta-9 Desaturase Intron1,4 of Gene can be linkage to controlled blood biochemical index traits and worthy of further studing as a molecular marker.SNPs genotypes of Delta-9 Desaturase Gene significantly effected the some fat traits. SNPs genotypes of Intron 1 significantly effected Intramuscular fat band width,sebum,sebum rate. SNPs genotypes of exon 4 significantly effected Intramuscular fat band width,sebum,sebum rate. SNPs genotypes of exon 4 significantly effected Heart weight,Head rate.The For Intron1,2,4, genotypes TT,GG,AA have a positive correlation with these traits. The results of this study suggest Intron1,2,4 of Delta-9 Desaturase Gene can be linkage to controlled fat traits and worthy of further studing as a molecular marker.SNPs genotypes of Delta-9 Desaturase Gene significantly effected the some fatty acid contents. SNPs genotypes of Intron 1 significantly effected C12:0,C14:0,C15:0,C16:0,C16:1,C18:1,C20:2,C22:6. SNPs genotypes of exon 4 significantly effected C16:2.The For Intron1,4, genotypes TT,AA have a positive correlation with these traits. The results of this study suggest Intron1,4 of Delta-9 Desaturase Gene can be linkage to controlled fatty acid contents and worthy of further studing as a molecular marker.
