Random amplified polymorphic DNA (RAPD) technique was applied to assess the genetic variations among inter- and intra-population of four wild stocks (20 individuals) of Macrobrachium nipponense , which were collected from Anhui Liuan, Anhui Longganhu Lake, Anhui Huainan and Jiangsu Gaochun. Under predetermined optimal reaction conditions, the amplifications gave 173 reproducible and stable fragments ranging from 200bp to 2500bp using 16 random primers selected from OPP and OPX 40 ones. There were 75 polymorphic fragments. Except OPX-18,OPP-09, there were all polymorphic primers(The proportion of polymorphic primers was 87.5%).Based on the RAPD data, the Shannon's information index of intra-population for Huainan, Gaochun, Liu'an, Longganhu were 0.2466 ,0.2089 , 0.1896 and 0.1771. Nei's genetic diversity of Macrobrachium nipponense was that Huainan population was the maximum, the Gaochun population followed ,the minimum was the Liuan population. The genetic distance revealed that the differentiation had taken place to some extend among the Macmbrachium nipponensis. The maximum genetic distance occurred between the stock of Gaochun and Longganhu populations(The number was 0.1712), and the minimum occurred between Huainan and Liu'an populations(The number was 0.1146). With the method of UPGMA on the basis of genetic distances, the results showed the populations of Liu'an and Huainan assembled one branch first, the last did Gaochun and Longganhu because of the maximum genetic distance Both results were accordant.
