| Dazu black goat is a group of goats that have excellent hereditary trait in Dazu county in Chongqing. The goat has big body form with pure black hair and can grow fast with good meat output. It has an especially high efficiency in procreation, which is about 350%400%. At present, breed selection is being done in this group and a breeding center of Dazu black goat has been founded locally. A nuclear herd of black goat was formed but the pedigree is not clear between individuals and this is disadvantageous to breeding. This study analyzed the relation of the goats in nuclear group using microsatellite DNA technology and it could be referred in establishing of a rational breeding system to avoid trait degeneration caused by inbreeding and to quicken the process of breeding.The object of this study is genome DNA of black goats. By jugular vein blood sampling we got 133 samples which included 104 offspring , 11 male stud goats and 18 mothers. According to the record, 24 offspring have known mother and the other 80 offspring's mother were unknown. After detection, the genome DNA extracted by phenol—chloroform method had fulfilled the quality request which was needed in this study .The loci used in the study were obtained from database via internet and loci could accord with the selection criterion were chosen. After synthesization of primer, temperature gradient was set and Mg2+ concentration was adjusted in order to optimize the PCR amplification conditions. At last 24 microsatllite loci were used in our experiment for analysis. The products of PCR amplification were detected by non-denaturalized polyacrylamide gel electrophoresis and the gels were dyed by silver nitrate. Whereafter gel images were obtained by scanning, analytical software Bandscan and pBR322 DNA/Msp I Markers were used to obtained the fragment size of loci amplification products, then the genotype database include 24 loci allele information for each individual was established.A special parentage analysis software Cervus2.0 was used for correlative statistical data processing and paternity analysis based on genotype database and relation record.
