Genetic diversity within and among populations of Qiongzhuea tumidinoda, Qiongzhuea macrophylla and Chimonobambusa rivularis were analyzed using RAPD markers. Through polymorphic genome analysis, 13 random primers for Chimonobambusa rivularis, were selected which producted distinct bands, showing unmistakable polymorphism. By NTSYS-PC program Nei's genetic similarity coefficients were generated and dendrogram was constructed using UMPGA. Basing on this analysis, we discussed the value and strategy of rare and endangered ornamental bamboos in biology diversity conservation, put forward countermeasure and suggestion of three rare and endangered ornamental bamboos, and those applied landscaping planning.This experiment results were follows:1. Optimized results of amplification reaction program and system: Amplification reaction program of three bamboos: After initial denaturation for 5min at 94 ℃,Then the reaction was continued for another 45 cycles through the following procedure:94 ℃ for 1 min, 36℃ for 1 min and 72℃ for 1 min30sec.Finally,it ended at 72 ℃ for 10min.Amplification reaction system for RAPD analysis: 10×Taq polymerase corresponding buffer(2μL), 1.25U Taq polymerase, 75ng template DNA, 45ng primer, 1.88mmol · L-1 MgCl2, 0.19 mmol · L-1 dNTP in total 20μL reaction volume.2. The analysis of amplification results of random primers for three bamboos' polymorphism shows:53 individuals in 4 populations of Qiongzhuea tumidinoda,13 primers amplified 254 bands, among which 236 were polymorphic attaining a polymorphic rate of 92.91%.26 individuals in 3 populations of Qiongzhuea macrophylla, amplifiing 250 bands, among which 236 were polymorphic attaining a polymorphic rate of 94.40 % .24 individuals in 4 populations of Chimonobambusa rivularis, 22 primers amplified 375 bands, among which 350 were polymorphic attaining a polymorphic rate of 93.33%.3. Three bamboos populations'genetic diversity analyzed by percentage of polymorphic loci (PPB): Compared with two Qiongzhuea tumidinoda populations 'genetic diversity, Mabian dafengdings' is little higher than Yibin junlians'; In Mabian dafengding,the high-low sequence of genetic diversity of three Qiongzhuea tumidinoda subpopulations:Mabian dafengding2(the average altitude: 1750m) > Mabian dafengding 1(the average altitude: 1654m) = Mabian dafengding3(the average altitude: 1839m) Mabian dafengdings' the high-low sequence of genetic diversity of three Qiongzhuea macrophylla subpopulations: Mabian dafengding3(the average altitude:2120m) > Mabian dafengding2(the average altitude:2030m) > Mabian dafengding1 (average altitude: 1915m). Qionglai tiantans'high-low sequence of genetic diversity of four Chimonobambusa rivularis subpopulations: Wu tonggang2(the average altitude: 1085m) >Wu tonggang l(the average altitude: 1022m) > Zheng tiantai2(average altitude: 1320m). > Zheng tiantai1(the average altitude: 1270m).4. Two bamboos populations' relationships analyzed by genetic similarity coefficients: Qiongzhuea tumidinoda and Qiongzhuea macrophylla populations 'genetic similarity coefficients was 0.6400.
