Effects of amino acids and vitamins on the in vitro development of buffalo embryos derived from parthenogenetic activation(PA) or in vitro fertilization( IVF) were investigated, and the effects of the mineral oil sources on the in vitro development of buffalo embryos derived from parthenogenetic activation(PA) were also examined in this study.Addition of 1.5% MEM nonessential amino acid solution (NEAA) to SOF medium resulted in a significant increase in the cleavage rate of either PA (79.8% vs 67.7%, P<0.05) or IVF embryos (65.7% vs 51.9%, .P<0.05). However, supplementation with 3.0% BME essential amino acid solution (EAA) decreased the cleavage rate of IVF embryos (42.2% vs 52.2%, P<0.05), although the hatched rate of PA blastocysts was increased by supplementation with 1.0% EAA (77.3% vs 43.3%, P<0.05). Addition of 1.0% MEM vitamin solution (Vit) to SOF medium increased the cleavage rate of IVF embryos (65.6% vs 49.4%, P<0.05)> but decreased the proportion of PA embryos developed to blastocysts (13.9% vs 26.7%, P<0.05) and hatched blastocysts (28.0% vs 50.0%, P<0.05). Significantly more PA embryos cultured with Sigma filterd oil or Sigma embryo tested oil developed to blastocysts on Day 7 (39.9% or 34.4%) and by Day 9 (42.8% or 40.2%) in comparison with Ameresco mineral oil (22.9% and 28.8%, P<0.05).In conclusions, (1) nonessential animo acids can stimulate the development of buffalo embryos derived from either PA or IVF; (2) Low concentration of essential animo acids is beneficial to the development of buffalo embryos but high concentration of EAA is detrimental to the development of buffalo embryos; (3) Vitamins are beneficial to buffalo IVF embryos but detrimental to PA embryos; (4) The in vitro development of buffalo embryos was influenced by the quality of mineral oil and the mineral oil from Sigma is suitable to embryo in vitro culture.
