Mature zygotic seeds and seed plant of the masson pine (Pinus massoniana Lamb) provenience from Guangxi tongmian were cultured as the initiation explants in order to investigate the procedures of sterilization, induction, subculture, elongation, roots development and transplantation. The result has been shown as follows:After the mature zygotic seeds of the masson pine were sterilized twice, its sterlization and germination rate was 69.7% and 67.9% respectively. Its germination rate was 71.3% under the natural environment. When the seed terminal buds of Masson pine that had been planted for 50 days were sterilized, the sterilization and survival rate both were 100%.The bud multiplication system has been established by the terminal bud. The induction rate is 100%, the multiplication coefficient is 4.95. The best age for the buds induction is about 100 days. The optimization medium is modified GD supplemented with BA3.0 mg/L and NAA0.05 mg/L . The optimization medium for the first subculture is modified 2/3GD supplemented with BA3.0 mg/L and NAA0.05 mg/L. It's induction and multiplication coefficient were 93.8% and 4.69 respectively. When they were surcultureed on DCR or 1/2 GD modified medium both supplemented with BA2.0 mg/L and NAA0.05 mg/L, their induction and multiplication coefficient were 100% and 3.07 and 90.6% and 4.22 respectively.The optimization medium for elongation of the induced bud is modified GD supplemented with 0.3% AC in which a bud with 0.6 cm could grow 5.2 cm high aider two months.The optimization medium for rhizogenesis is modified 2/3GD supplemented with IBA 4.0mg/L, the rooting rate is 26.7%.The seeding by tissue culture was transplanted into the medium of sursoil disinfected by 0.1% KMnO4, and it's survive rate was 100% after one month.
