Transformed hairy roots of Polygonum multiflorum Thunb. were obtained by the infection of Agrobacterium rhizogenes strains LBA9402 and the suspension culture systems of the hairy roots were established; some ingredients were analysed and some were isolated firstly from the hairy root and identified ; and the fundermental experiment of biotransformation was carried out successfully.The new hairy root system was established, the growth cycles of which reduced from 40 days to 30 days. In one growth cylce the growth times increased from 11.03 to 42.09, and the anthraquinones yields increased from 16.91 μg/flask to 41.95 μg/flask. The growth curves and accumulating curves of anthraquinones were established.TLC and HPLC proved that emodin and rhein were existed in the hairy root with free and combinative forms. Five compounds were obtained by application of column chromatography and sephadex HL-20, and part of their structures were determined by physical and chemical properties, as well as spectrographical identification. Two of them are β-sitosterol and β-sitosterol-3-O-β-D-glucopyranoside. All of these compounds were isolated firstly from hairy roots of Polygonum multiflorum Thunb.The fundermental experiment of biotransformation was carried out with the fastest-growth hairy root and the TLC and HPLC confirmed that andrographolide was biotransformed successfully.
