Polygonum Multiflorum Thunb is perennial herbaceous vine and one of the most important and traditional herbal drugs in China, which is prescribed for the dried rhizome and root of Polygonum Multiflorum Thunb of the family Polygonaceae. Because of the high nutritional and medicinal value, it receives people's favor increasingly. However, Due to the over collection, feral medicinal material is destroyed. At present, people are conducting on artificial cultivation in some area, so a good variety is very important. Plant polyploidy generally has strong adverse-resistance, good economic character and high yield. Polyploidy breeding has always been a main field of plant breeding. Someone find triploid plant of Polygonum Multiflorum Thunb. And it's a good material for studying good variety of Polygonum Multiflorum Thunb. Therefore, it's important to study genetic relation of triploid and diploid in Polygonum Multiflorum Thunb, the reason to route to polyploidy and the origin of polyploidy.We investigated genetic variation of Polygonum Multiflorum Thunb with cytological markers and molecular markers. And the main results were as follows:1. Improved BSG method was used to study the C-banding of Polygonum Multiflorum Thunb from 7 different populations. The results of karyotypic study showed that it is also a triploid from Guangdong province Gaozhou population. And other populations are diploid except for Guangxi province jingxi triploid population. It explains that polyploidization is not individual phenomenon.There were many differences in the banding patterns among the populations, which included the different location and number of C-banding. The chromosome banding pattern was mainly consisted by telemetric band, centromeric band and intercalary band. All these results indicated that Polygonum Multiflorum Thunb was on a higher level of evolution system. Compared the three genomes of the triploid, most C-banding pattern of the homologous chromosomes are consistent.Furthermore, karyotype date was studied through statistical analysis. It's found karyotype resemblance-near coefficients among Guangdong province Gaozhou and Deqing populations, Guangxi province Jingxi and Deqing populations are the two largest values. It shows the two triploids are autotriploid. UPGMA clustering demonstrated that the consanguinity of the diploid and triploid are very close and clustered together early.2. The RAPD system suitable for Fallopia multiflora was developed through orthogonal design and single factor design experiment. With the technique of RAPD, 34 random primers which are fit for Polygonum Multiflorum Thunb RAPD study are screened from 240 random primers.3. Using 34 random primers, with the improved RAPD system, 10 populations of Polygonum Multiflorum Thunb were studied. The result shows the SM coefficient varied from 0.5881-0.8602. The analysis of SM coefficient and UPGMA clustering demonstrated that the samples were divided into five clades. The two triploids and Guangdong diploid populations were clustered together early. And other populations had its own clade. The result of the principal coordinates analysis is consistent with UPGMA clustering analysis.
