In China, Siniperca chuatsi had a relatively high market value, and was widely cultured throughout the country. However, outbreaks of diseases caused by viruses,bacteria and parasites have caused severe economic losses to the aquaculture industry, and in some cases the mortality can reach as high as 100%. Despite the economical importance of the fish and severe economic losses caused by diseases, little research has been carried out on the mandarin fish vaccine. Siniperca chuatsi virus and Aeromonas hydrophila are two of the most causative pathogenic agents to Siniperca chuatsi, and two pathogens often infect the fish together, so a bivalent vaccine which can prevent Siniperca chuatsi from both Siniperca chuatsi virus disease and Aeromonas hydrophila disease were urgent need for Siniperca chuatsi culture development. In this paper, we studied the immune dose and immunogenicity of three kinds of vaccines to Siniperca chuatsi, which were formalin-killed whole Aermonas hydrophila cell, lypopolysaccharid (LPS) of Aermonas hydrophila and Major capsid protein of Siniperca chuatsi virus (MCP), then a bivalent vaccine was produce with LPS and MCP, and the immune efficacy of the bivalent vaccine was studied.1. Copariation of Siniperca chuatsi immune response to whole cell Aermonas hydrophila and lypopolysaccharid of Aermonas hydrophila. In reference to immune dose of formalin-killed Aermonas hydrophila and Aermonas hydrophila LPS to other fishes, Siniperca chuatsi were intraperitoneal injected with formalin-killed Aermonas hydrophila and lypopolysaccharid of Aermonas hydrophila (LPS), Aermonas hydrophila were injected at dose of 1.0×10~8cfu/mL,3.0×10~8cfu/mL,6.0×10~8cfu/mL 0.2mL every fish and LPS at dose of 20ug/g,25ug/g and 30ug/g separately. Rssults showed antibody in Siniperca chuatsi serum to formalin-killed Aermonas hydrophila and Aermonas hydrophila LPS both arrived 128, but the relative survival to Aeromonas hydrophila challenge were 77.8% and 83.3% separately.2.
