Study On Tissue Culture Of Two Fine Clones Of Hippophae Rhamnoides L.

Wood Stems,hydroponics buds and leaves, as well as field condition leaves of"Ze liang"(Hippophae rhamnoides L.) and"Xiang yang"(Hippophae rhamnoides L.) were cultured on basic mediums MS, B5 and WPM , multiplication and some factors which influence culture process were also researched systematically. The main results as following: (First) Asepsis cultivation foundationWood stems are not suitable to culture as explants, because of high contamination rate and hard sterilization completely; hydroponics stem apexes are young and keeping low contamination rate, so it's a appropriate explant to set up asepsis cultivation. The best disinfectant time is alcohol marinating 40 s, HgCl2 marinating 3 ~ 5 min. (Second)Primary culture and subculture of stem apexesâ… . The primary culture medium of"Ze liang"stem apexes is MS +6-BA 0.5 ~1.0 mg/L +IAA 0.5 mg/L, explants grow fast and well, no significant difference among dormancy periods of the five months;Subculture medium is 1/2 MS +6-BA 0.5mg/L+IAA 0.3 mg/L+AC 0.2 %.â…¡."Xiang yang"primary culture medium chooses 1/2 WPM +6-BA 0.5 mg/L +NAA 0.2 mg/L or WPM +6-BA 0.5 mg/L +NAA 0.2 mg/L, the better explant collection times are November,Febrary and March, vitrification is not serious during these periods , it's no good of improving concentrations of sugar and agar to vitreous; subculture medium is 1/4 MS +6-BA 0.5 mg/L+IAA 0.3 mg/L.(Third) Axillary buds inductionAxillary buds can be induced from young stems, on MS +6-BA 3.0 mg/L +IAA 0.05 mg/L, an average of 1.53 axillary buds of"Ze liang"are obtained, 8.7 adventitious buds are induced from root stems. Young stems of"Xiang yang"can be induced 1.67 axillary buds, and 2.0 adventitious buds attained by root stems. (Fourth)Leaves cultureHydroponics leaves,field condition leaves and aseptic plant lesves of two clones'are all can be cultured on appropriate mediums.
