The efficient, stable plant regeneration system is a key to success for transformation. Factors affecting the high efficient regeneration of sea buckthorn were studied in vitro cultured somatic embryos, including plant growth regulation, the length of dark incubation, polarity of leaf, basal media, type of explants, LH. .hereby the high-frequency regeneration system of sea buckthorn was established. The main results were as follow as:1 It is the best somatic embryogenesis effect that explants on 1/2B5 medium ,containing Sugar 30g/L .1/2B5+6-BA3.0mg/L+NAA0.1mg/L is the optimum medium that induction of embryogenesis from the field leaves of hippophae rhamnbides L. subsp.sinensis Rousi; 1/2B5+6-BA2.0mg/L+NAA0.1mg/L is the optimum medium that induction of embryogenesis from the tube leaves and regenerative leaves . 1/2B5+6-BA0.5mg/L+NAA0.05mg/L is the optimum medium that Cotyledon induce somatic embryogenesis.2 Cotyledon dark incubation experiment result showed :For the cotyledon of hippophae rhamnbides L. subsp.sinensis Rousi, a period of dark incubation from 2 to 3 weeks was necessary to the high efficient regeneration system of somatic embryogenesis.3 Cotyledon polarity experiment result showed:it was noted that the close and far axial head both form somatic embryos ,but regeneration rate of close axial head prior to far axial head . Somatic embryo mostly form on the close axial surface.4 The cotyledon ,the hypocotyls ,the leaf ,the aseptic stem segment of training the condition to be the optimum ,the regeneration rate of the cotyledon was the highest.5 Medium of addition LH restrict regeneration of the cotyledon of hippophae rhamnbides L. subsp.sinensis Rousi6 With B5+6-BA0.05mg/L+NAA0.5mg/L medium, it had the best result in rooting.
