Hippophae rhamnoides L.has high values in economy and environment. It is both an important species to the development of economy and a better species for ecological construction. In order to meet the for excellent young plants and speed up the resources building of Hippophae rhamnoides L, Two elite Russia Hippophae rhamnoides L. cultivars"Ze liang"and"Xiang yang"were cultured. The culture techology of leaves, axillary buds, root induction, acclimatization and transplantation were optimized.The main results were as follow:(1),The suitable positions on the stem apexes and different parts of leaves were selection and established in leaves cultureThe suitable explant to induce callus is the middle and proximal parts of leaves from the upper and middle parts of the stem apexes for "Ze liang", the under-part and the middle parts of leaves were easier to induce adventitious shoot and there was no difference for different parts of leaves to induce adventitious shoot for "Ze liang", but it's better to choose the middle of leaves to induce adventitious shoot for different parts of leaves.For "Xiang yang", the upper leaves on stem is situable to induce callus, the middle leaves on stem is situable to induce adventitious shoot; The regeneration ability is different for different parts of leaves, and the middle of leaves is both siutable to induce callus and adventitious shoot.(2),Optimization and establishment the culture medium for leaves1/4 MS+6-BA 0.5 mg/L+NAA 0.04 mg/L is suitable for "Ze liang" to induce adventitious shoot, the average shoots is 6.3 and the adventitious shoot induction rate is 66.67 %.For "Xiang yang", on 1/2 WPM + 6-BA 0.6 mg/L + NAA 0.04 mg/L the the average shoots is 3.25 but the adventitious shoot induction rate is only 25 %, on 1/3 WPM + 6-BA 0.4 mg/L + NAA 0.02 mg/L the the average shoots is 2.23 and the adventitious shoot induction rate is highest,up to 56.67 %.(3),Optimization the culture medium and establishment superior of leavesFor "Ze liang", MS + 6-BA 3.0 mg/L +IAA 0.05 mg/L is the siutable medium to induce axillary buds from stem. The average axillary buds is 0.78, axillary buds induction rate is 37.5 %.For "Xiang yang", 1/2 WPM + 6-BA 4.0 mg/L +NAA 0.02 mg/L is siutable to induce axillary buds from stem, the average axillary buds is 0.83, axillary buds induction rate is 52.5 %.The axillary buds induction rate is higher when the leaves on the stem were cut partially.(4),Optimization and establishment the root culture medium for different variefies For "Ze liang", 1/2 B5+6-BA 0.15~0.18mg/L+IBA 0.2mg/L is siutable to induce root, the root induction rate is 90 %.For "Xiang yang", 1/2 B5+KT 0.5mg/L+NAA 0.03~0.05 mg/L is siutable to induce root, the root induction rate is 86.67 %.(5),Establishment the suitable technology for acclimatization and transplantationIt's easier for tissue-cultured plantlets to survive when perlite /turives is 2:3, pH value is 7.0, at the same time the humidity in the air and medium moisture must be keeping in a suitable level, the survival rate is up to 66.67 % and plantlets grow well.
