This paper summarized the purpose and the signification of the research has been made about the actinomycetes acquired from the saline soil. Introduced the research development and the application foreground of isolate and classify about extreme microorganism, especially the halophilic actinomycetes. In this thesis, we emphasized the study status of actinomycetes taxology, the study messure of polyphasic taxanomy, the research character and the development current. The same time, we introduced the microorganism molecular ecology, especially the keystone, common method, and the application in practicality of culture independent technique.This thesis was aimed at the saline soil collecting from Ningxia. Isolated and choose the actinomycetes from it. We discussed the influence complication of actinomycetes isolate effect from different culture media, filtered the strains from this acquired actinomycetes and conserved in our lab with the character of special form, tolerant the salt, and secrete special antibacterial substance. The actinomycetes which have been isolated were classed by the polyphasic taxonomy technique including form character, physiology and Biochemistry determine, chemistry component analysis and the comparability contrast of 16SrRNA gene sequence. The classified status was confirmed from three different aspect including phenotype, gene type and system develop. Simultaneity, the technique of culture independent was attempt for the diversity analysis of microbial acquired from saline soil environment. The research results indicated:1. The number of actinomycetes was improved by design the culture different from media components, the type of salt ion and the salt concentration. We posted the number, category and distribution rule of the actinomycetes from representative saline soil in Ningxia.2. Sixty strains and tolerant 18% NaCl concentration strain was acquired according as special form, high saline tolerance and produce the antimicrobial substance controlling the pathogeny development in agriculture. This offer a good primary strains for the discovery of rare strains and the application of function strains.3. The classified status of test strains were confirmed by the polyphasic taxonomy technique including morphological characteristics, culture characteristics, physiological characteristics, and cell chemistrical component analysis and the comparability contrast of 16SrDNA gene sequence.
