The paper could be divided into two parts. In first part, A high performa-nce liquid ch-romatographic method using fluorescence detection was developedfor simultan-eous separation and determination of 8 quinolones, Pipemidic acid,Ofloxacin, Ciprofloxacin, Danofloxacin, Enrofloxacin, Sarafloxacin,Oxolinic acid,Flmequine.Linearity was verifieh in range from 30 to 2000μg/L, the correlationcoefficient was better than 0.999. The limit of quanification(LOQ) was 30μg/L,the limit ofdetection (LOD) was 5μg/L (Pipemidic acid:20μg/L). The analytes were extract-ed by dispersing samples and microwave extraction. This method was suitablefor fish muscle, chicken muscle, chicken liver and pig kidney, the re-coveries were70.0-99.5%, RSD:1.0-8.5%. Compared with SPE (RPS,HLB and MAX cartridges), this method was better. Different responsibilities of 8 quino-lones in photodiode-array and fluorescence detection were descuibed. The effectsof mobile phase pH value and composition on responses and separation of 8 quinolones were investigated. Chromatographic conditions were optimized. Thecolumn used was Aglient XDB-C18 (5μm,150mm×4.6mm i.d.),with phospho-ric acid:water:trie-thylamine:acetonitrile as mobile phase. The excitation and emi-ssion wavelengthswere 285nm and 460nm for Pipemidic acid, Ofloxacin, Cipro-floxacin, Danoflo-xacin, Enrofloxacin, Sarafloxacin; 325nm and 365nm for Oxo-linic acid,Flmequi-ne, respectively. The uncertainty in measurement was 6.1-5.9.This method wassensitive, accurate, simple, rapid. In second part, the pharmacokinetics properties and residue depletion of ox-olinic acid and flumequine were studied after vascular administration to Mur-a...
