There are two major questions of produce system of papaya propagation in present. One is higher cost, the other is lacking of a suitable and effective monitor system for identification to plantlets'quality. There are two advantages to establish the technical platform/foundation of industrialized produce and quality monitor system of papaya propagating plantlets ,one is that it can establish a better foundation for producing anti-virus seedlings of papaya, the other is it can greatly promote the process of tissue-culture papaya industrialization.The papaya propagation system was optimized succeedly in this study at some aspects. Using the cheap edible sugar as carbon source instead of sucrose in culture medium, the result showed that the propagation coefficient and fresh weight increment kept equivalent compared with the pre-system, and the optimized weight of sugar was 38g/L and the optimum value of kara gel as solid holder in culture medium was 5g/L;The result of cutting experiment showed that the livabilityof plantlets could reach 32%,when marinating the non-root propagating plantlets in 1mg/L IAA for 3hr; The result of optimized transplant showed that the livability could get 80% when treated the seedlingbed being dealed with formaldehyde(dilute1000 times)and DiJunLing(dilute 1000 times),and with membrane being covered for two days,the plantlets were marinated 3h with 1mg/L IAA before their being transplanted .Studying the genetic stability of the tissue-culture plantlets by cytologic method, it was found that the chromosomes were not integral after shoots being subcultured in 32 generation continuously , and the fructiferous rate and anti-virus character were droped when the plantlets were planted in field.Whether the tissue-culture seedlings were infected by virus and their ability of anti-virus were detected by biology and serology and molecule biology method, and it was found that all three strains were non-virus; and the no.2 strain had the strongest anti-virus ability, the no.3 and no.1 strains were next.
