Jining Blue goat with high prolificacy is an exceptional local goat breed in China. Bone morphogenetic protein 15 (BMP15) gene which controls fecundity of Inverdale, Hanna, Lacaune, Belclare, Cambridge and Small Tailed Han ewes was studied as a candidate gene for the prolificacy of Jining Blue goats, to provide scientific evidence for the basic genetic research on high productivity of Jining Blue goat.Both V31D mutation in Inverdale ewes and Q23Ter mutation in Hanna ewes of BMP15 gene were not detected in Jining Blue goat, Liaoning Cashmere goat, Inner Mongolian goat and Boer goat by PCR-RFLP. The results indicated that maybe the fecundity of Jining Blue goat was not affected by both V31D mutation and Q23Ter mutation of BMP15 gene,.According to the sequence of ovine BMP15 gene, six pairs of primer were designed to detect single nucleotide polymorphism in exon 1 and exon 2 of the BMP15 gene in both high fecundity breed (Jining Blue goat) and low fecundity breeds (Liaoning Cashmere goat, Inner Mongolia Cashmere goat, Boer goat) by PCR-SSCP. Only the products amplified by primer F5/R5 displayed polymorphism. Results indicated that two genotypes (AA, AB) were detected in prolific Jining Blue goats, and only one genotype (AA) was detected in low fecundity goat breeds. In Jining Blue goats, frequency of genotype AA and AB was 0.10 and 0.90, respectively. Sequencing revealed two point mutations (G963A and G1050C) of BMP15 gene in the AB genotype in comparison to the AA genotype. The genotype distribution was significantly different between prolific Jining Blue goats and low fecundity goat breeds (P<0.001).
