Bone morphogenetic protein 15 gene is the major gene for fecundity of Belclare and Cambridge ewes,and a single amino acid change in BMP 15 protein directly impacts ovarian follicular development and ovulation in sheep.The polymorphism of BMP15 gene was detected in Guizhou White goat by restriction fragment length polymorphism(R.FLP) and single-strand conformational polymorphism(SSCP) analysis.The results indicate that:1.The DNA fragments of exon 2 with 842bp in BMP15 gene was cloned taking the genomic DNA as templates extracted from Guizhou White goat.By comparing to Jining qing goat,sheep,cattle,pig,dog,human and mouse,the similarity of deduced amino acid (AA) sequence were 99%,99%,97%,90%,86%,81%and 80%respectively.It suggested that the BMP 15 gene is conserved in mammals.2.The polymorphism of BMP 15 gene was detected in the white goat of Guizhou by RFLP and SSCP analysis.The results showed that the mutation of FecXG was not detected in all samples,and FecX~B mutation was found in female goats with triplets and male goats. Sequencing results confirmed that the FecX~B genotype contain a mutation at 1100bp(G/T) in the coding region of BMP 15 gene.The G/T mutation caused the substitution of serine to isoleucine at residue 367 of the coding region for mature peptide(S99I).3.FecX~B mutation was only found in female goats with triplets and male goats,and all of the FecX~B mutation was identified in heterozygous genotypes(AB).The mutant was defined as AB genotype(4.2%),and the ordinary individuals were AA genotype(95.8%),frequency of allele A(97.9%) was much higher than allele B(2.1%).4.The entire coding region for BMP15 mature peptide was detected by SSCP analysis in Guizhou White goat.All of the mutant sites including FecX~I,FecX~H and FecX~L were not found and there did not found any other new mutant in BMP15 gene except for FecX~B.It suggested that FecX~B mutation is related to the regulation of fecundity in Guizhou white goat.
