With the materials of leaves,epicarp and seeds of pomegranate coming from xinjiang hetian piyaman, the experiment showed that the total flavone content in pomegranate epicarp was higher than that in others. Then measured the content of rutin, quercetin, kaempferol in leaves,epicarp and seed of pomegranate with HPLC. In addition, analyzed effects of concentration of alcohol, the temperature of extracts, the time of extracts, the ratio of liquid and solid the time of ultrasonic treat on the flavone extraction ratio by orthogonal test. Moreover, using the method of DPPH-Microplate and the antioxidative activities in lard estimated the antioxidant activities of bioactive products in leaf and epicarp of pomegranate. Finally, the experiment studied the effect of flavone compounds on staphylococcus aureus and colibacillus in order to provide theory basis for using the resource.The major results can be summarized as below:1) Qualitative test: the extractor of leaves and epicarp and seed of pomegranate, the peachblow foam produced by oxidation-reduction reaction, the green brown substance come by phenol–hydroxide reaction, by complexation reaction kelly fluorescence appeared, the phenomenon in different condition show the compound of flavone in leaves and epicarp and seed of pomegranate.2) Quantitative test: with Ultraviolet spectrophotometry, the amount of flavone in epicarp was 5.804%, in leaf was 4.622% and in pomegranate's seed was 1.056%. With HPLC there was rutin 1936.24 mg/kg, kaempferol 0.211 mg/kg and no quercetin in pomegranate's seed and leaf, none in epicarp.3) The test of the alcohol's concentration show that the ratio of extract flavone was best in 70% alcohol, no better than in 60% which was better than in 40% , 50% , 80% , 90% .4) The test of temperature show that the ratio of extract flavone increased with the temperature rised, however the ratio of extracted flavone was down in 80℃.5) The test of time to the flavone extracted from the leaves of pomegranate show that in general the longer time, the more flavone, if the extracted time between one or two and half hours. while the diffuse speed was insignificant the extract speed if the time over two and half hours.6) The test of the ratio of solid and liquid show that the influential factor was the ratio of solid and liquid. There were insignificant differences of the ratio of extraction flavone when the ratio of solid and liquid between 1:15 and 1:10. 7) Ultrasonic treatment on leaf the ratio of extracted flavone was significant. The highest ratio of extracted flavone, 4.72% when the leaves were treated half-hour, while this way can save quintupling more times.8) Orthogonal test with ultrasonic extraction show the best parameter was 60% alcohol, and ultrasonic extract temperature 70℃, the ratio of solid and liquid was 1:15.9) The test of bacteria inhibitation in vitro show: the extraction from epicarp and leaf of pomegranate have the antifungal activity to staphylococcus aureus and colibacillus, the epicarp minimal concentration of the bacteria inhibitation to staphylococcus aureus was 0.781mg·mL-1, to colibacillus was 3.125mg·mL-1, the leaf minimal concentration of the bacteria inhibitation to staphylococcus aureus was 12.5mg·mL-1, to colibacillus was 25mg·mL-1.10) DPPH·Microplate assay and the antioxidative activities in lard was made to estimate the antioxidant activities of bioactive products in epicarp and leaf of pomegranate, the result show the free radical scavenging activity of extractor with alcohol from leaf was higher than the extractor from epicarp. Results show the leaf was 93.35%, the epicarp was 80.65%, the extractor has antioxidative activity in lard.
