Cortex Magnoliae Officinalis(CMO),as a Chinese traditional medicine,originated from the dried cortex of trunk, branch and root of Magnolia officinalis Rehd. et Wils. or M. officinalis Rehd. et Wils var. biloba Rehd. et Wils. Resource status, effective components and HPLC fingerprints of CMO were studied systematically to offer referrence for evaluating CMO scientifically according to the field survey of the five national production bases,along with information correlative recent years. It is the first time to compare HPLC fingerprints of CMO of different ages and sampling parts, and so do the effective components and HPLC fingerprints of original and regenerate plants.CMO is rare and endangered in our country. The wild resource is approximately disappeared in the investigation and to strengthen the protection of the wild resource is imminent.The samples were extracted with ultrasonic in methanol, and the effective components content was measured by HPLC. The chemical compositions were studied by the hierarchical clustering analysis and discriminant analysis to conduct the chemical pattern recognition. The samples were distinguished and classified quickly and correctly, and the samples from Hubei and Sichuan were better than others.To analysis the effective components of CMO of different ages,sampling parts, original and regenerate plants in Enshi Hubei. The results indicated that: the content of the effective components was rising according to the ages, and arrived to the maximum in 21 years around; the content in root was the highest, in trunk was the next and in branch was the worst; there were no significant differences between original and regenerate plants.The main factors influencing the quality of CMO were studied as well. It was that the quality was better if the circumference was bigger and the color of powder was darker.The extraction and chromatogram methodology were researched systematically. That was extracting with ultrasonic in methanol about 30min, with methanol-water-acetic acid(75: 23.5: 1.5) as the mobile phase ,UV detection at 254nm ,the flow rate of 0.8 mL·min-1 and clocked 60min.The HPLC fingerprints of different producing areas were studied by the hierarchical clustering analysis and common pattern was set up. The similarities were figured out and the plants in Hubei and Sichuan had higher similarity than others which was consistent with hierarchical clustering analysis.HPLC fingerprints of phenolic constituents of CMO of different ages, sampling parts, original and regenerate plants in Enshi Hubei were studied as well. According to the comparison of similarities, it showed that the quality was advanced with ages; trunk and branch had higher similarities with common pattern, but root was lower; there were no significant differences between original and regenerate plants.
