The objectives of this study were to investigate the effect of different extenders and taurine supplementation in different extenders on quality parameters and IVF of liquid rabbit semen . Three experiment were conducted.Experiment 1: The effect of different extenders on quality parameters of liquid rabbit semen were studied. The sperm mobility, percentage of swollen sperm, percentage Intact-acrosome sperm, sperm forward progressive, progressive speed and path velocity in A and B for 72h were significantly higher (P<0.05) than that extenders in C, D and E.. Extenders in A and B were better than extenders C, D and E.Experiment 2: The effect of taurine supplementation in different extenders on quality parameters of liquid rabbit semen were studied. Extenders in A and B were used as a foundation liquid, the sperm mobility, percentage of swollen sperm and percentage Intact-acrosome sperm in 10mM taurine treatment groups was significantly higher(P<0.05) than that of control for 24 h, 48 h and 72 h. The sperm progressive speed and path velocity in groups of 10mM taurine treatment shows no significant difference(P<0.05) for 24h, but significantly higher than that of control for 48h and 72h. When extenders in A and B were used as a foundation liquid, the taurine supplementation in liquid rabbit semen of the more ideal density be a 10mM.10mM taurine supplementation in A and B have a better protection function to sperm membrane and acrosome membrane, and can also sublimate sperm mobility.Experiment 3: The results of IVF in the fresh semen and the semen that stored for three days in B and taurine supplementation in B were researched. When carry out IVF in the semen that stored for three days in A/B and taurine supplementation in A/B, the cleavage rate and blastocyst rates of 10mM taurine treat groups(P<0.05) were no significant difference than that of control and other treat groups, but the cleavage rate and blastocyst rates of fresh semen were significantly higher(P<0.05) than that of control. Thus, 10mM taurine supplementation in rabbit liquid semen can elevate the cleavage rate and blastocyst rates during IVF.The quality parameters of extenders in A and B which taurine supplementation were higher than that of control significantly, but the groups of 10mM taurine treatment were higher than control when fertilized in vitro, and have no significant difference between them, the results of this need further study.
