Artificial insemination of livestock is an important part of efficient production in animal husbandry,and the cryopreservation of semen is essential for the widespread application.In Rex Rabbit breeding industry,there is no report on semen cryopreservation of chinchilla rex rabbit at present.In order to obtain the most effective storage conditions and methods for blue and purple blue rex rabbit semen,with a view to improving the utilization rate of the breed male rabbit and the reproductive performance of the female rabbit,thereby providing technical support for reducing production costs,this experiment studied the effects of different preservation methods(different dilutions,pH values and temperatures)on the sperm quality and artificial insemination offspring.In this study,we used 0.9% normal saline diluent ?,glucose-sodium citrate-EDTA diluent ?,glucose-sodium citrate diluent ? and glucose-citric acid-Tris-DMSO diluent ? to preserve the semen of chinchilla rex rabbit under the conditions of normal temperature(17?),low temperature(4?)and freezing(-196?),respectively.The main results are as follows: Under normal temperature preservation,diluent ?(Tris,glucose,citric acid,etc.)as the base solution and in the range of pH 6.3 to 6.7,the preservation solution with the optimal pH(6.5)was superior to the other three diluents in terms of preservation effect.The sperm viability of diluent IV-1 group(63.33%)was significantly higher than that of other diluents combinations(P< 0.01)at 48 h,and that of pH 6.5 group(65.01%)at 60 h was significantly higher than that of other groups(P < 0.05).In the study of low temperature preservation,the combination of diluent IV-2(Tris,glucose,citric acid,etc.)with pH value of 6.5 was superior to the other three diluents,and the sperm viability(48.42%),plasma membrane integrity rate(21.03%)and acrosome integrity rate(87.58%)of ?-2 were significantly higher than those of II-2 and ?-2(P < 0.01)at 72 h.For cryopreservation,the sperm viability(30.64%)and acrosome integrity rate(38.29%)were significantly higher in the combination of diluent ?(Tris,glucose,citric acid,etc.)and pH 6.5(P < 0.01)with respective to other groups.In the experiment of artificial insemination,the semen of chinchilla rex rabbit was preserved for 48 h with diluent ?(Tris,glucose,citric acid,etc.)as the base solution.Compared with fresh semen group,the effect of semen preserved at normal temperature(7.12)(P>0.05)was the best,followed by semen preserved at low temperature(7.10)(P>0.05)and frozen semen(6.75)(P<0.01)when considering the the number of live birth as the test index.
