Study On The Immunoregulatory Effects Of Compound Radix Astragali Preparation(CRAP) In Mice Using Seropharmacology Method

Compound Radix Astragali preparation(CRAP) was composed by Radix Astragali, Rhizoma Smilacis Glabrae, Herba Houttuyniae and Angelica. Experiments were carried to study its mechanism. Firstly, the prepare method of medicine serum and the amount of serum addition of CRAP were studied to explore CRAP serum pharmacology method. Secondly, we explored the pharmacology and immunoregulatory mechanism of CRAP, the proliferation of lymphocytes as well as the production of IL-2 were detected.In this experiment, three dosage groups were set up as follow: lower dosage group, moderate dosage group and higher dosage group. CRAP was introduced into mice through gastric route in one day, three days and six days. MTT assay was used to observe the influences of medicine serum with different dose, different drug times to the proliferation of spleen lymphocytes, for the purpose of confirming the optimal making method of medicine serum. The results indicated that: In one day experiment, the lower, moderate and higher dosage could improve the proliferation of spleen lymphocytes, comparing to the control group, but there were no significant differences. In three days experiment, the moderate and higher dosage groups could improve the proliferation of spleen lymphocytes with significant differences (P<0.01). In six days experiment, the moderate dosage group could improve the proliferation of spleen lymphocytes with significant differences (P<0.01) to control group. At the same time, for the moderate group, significant difference was observed when comparing the three days experiment to six days experiment. So the optimal dosage was 25g/kg of body weight and the optimal time of pouring medicine was six days.After confirming the optimal prepare method of medicine serum, we explored the amount of serum addition in the in vitro experiments. Two experimental groups were set up, one was control group, with no-drug serum were included; the other was experimental group, which included drug serum.
