IGFs was inslin-like polytides which stimulated cell proliferation and differatiation. It included IGFs (IGF-I and IGF-II), IGFR, IGFBPs. These were important functions in mammary gland development.IGFs was important in mammary gland development and lactation. Knowing about the functions and regulation of IGFs in mammary gland would help Knowing the mechanism of mammary gland development and lactation and find the way to stimulate the development and lactation of mammary gland and increase the production of milk in these research, we investigated the impact of IGFs on expression of IGFBP-5 and cyclin D1 systematically and the development of pregnancy mammary gland by explants cultured in vitro by culturing the mammary epithelial cells of mouse with IGFs in medium, we detected the apoptosis by fluorescence microscope.these illuminate the impact of IGF-I and IGF-II on expression of IGFBP-5,cyclin Dl and the impact of IGFs in the development of mouse mammary gland and impact of IGFs in apoptosis of lactation mouse mammary. These could provide the new way and theory for controlling the milk production and quality, promote the development of animal husbandry. It has important theoretical and practical significance.The results showed that IGF-I,IGF-II affected the expression of IGFBP5 in virgin, pregnancy,lactation involution and forcing involution mammary gland. Both IGF-I and IGF-II inhibited the expression of IGFBP-5. But the impact of IGF-I was stronger than that of IGF-II. The inhibition of IGF-I and IGF-II were not obviously (P>0.05) in virgin and pregnancy. In lactation, involution and forcing involution, IGF-I and IGF-II inhibited the expression of IGFBP5 obviously. (P<0.05)oIGF-I and IGF-II also affected the expression of cyclinD1 in virgin,pregnancy, lactation, involutionand forcing involution mammary gland. Both IGF-I and IGG-II simulated the expression of cyclinDl, But the impact of IGF-I was stronger than that of IGF-II in virgin, pregnancy, involution and forcing involutionmammary gland, IGF-I and IGF-II stimulated the expression of cyclin Dl obviously (P<0.05). The stimulation of IGF-I and IGF-II to cyclin Dl were not obviously (P>0.05) .Both IGF-I and IGF-II could promote the development of mouse mammary gland, the impact of IGF-I was stronger than that of IGF-II, but this was not obviously.Both IGF-I and IGF-II inhibited the apoptosis of mammary epithelial cells in lactation mammary...
