IGFBPs in circulation system of the animal organisms can bind the IGFs, regulate IGFs half-life period and enhance IGFs biological activity. IGFBPs which secreted from liver and ambitus tissues can regulate and stimulate growth of tissues. In this study, Nanjiang yellow goat was used as experimental material. Total RNA was extracted from liver of Nanjiang yellow goat at the 0th after birth, then the complete CDS of IGFBP-1and IGFBP-5 genes were cloned by RT-PCR.The characterization of expression of IGFBP-1 and IGFBP-5 genes in different genders, different tissues and different growth periods by real-time PCR (SYBR greenâ… ) method. The main results were showed as follows.(1) The sequence lengths of IGFBP-1and IGFBP-5 were 792bp and 816bp, respectively, which coded 263 and 272 amino acid residues. The IGFBP-1 nucletide and amino acid of goat and sheep were 99% and 98% identity, respectively.The IGFBP-5 nucletide and amino acid between goat and sheep were 98% and 99% identity, respectively.(2) The bioinformatics analysis indicated that IGFBP-1 protein maybe a hydrophobicity protein, but IGFBP-5 protein maybe a drophilic protein. IGFBP-1 and IGFBP-5 protein all had two transmembrane domains, signal peptide, the phosphorylation and the glycosylation. There wre three kinds of secondary structure in goat IGFBP-1and IGFBP-5 protein.The proportion of random coil was the highest one, the next was alpha helix, and the beta fold was the lowest. Phylogenetic analysis indicated that the goat IGFBP-1 and IGFBP-5 protein had high homology with other mammal.(3) The expression level of the two genes in liver was higher than in other tissuses,such as heart, spleen, lung and muscle tissues(0.01
