Roe deer is widely distributed in the Northeast of China as an important animal. The population in the wild, however, has declined significantly due to excessively hunting in resent years. Therefore, understanding the genetic diversity and population's hereditary structure of the Roe deer, and protecting the limited wild Roe deer resource well are urgent tasks.With the development of molecular biology, genetic marker technique is a rather accurate method in molecule level to study hereditary variation in wild animals. There are few reports about the wild Roe deer's genetic marker study in literature, and we have not found any reports about hereditary variation studies of Roe deer in northeast of China in molecule level. For the sake of the Chinese northeast area in primary understanding Roe deer's hereditary structure, Nine community of Roe deer that this research collected Chinese northeast region three areas department, apply segmental sequence variation of mitochondrial DNA (mtDNA) control the area and genetic marker of random amplified microsatellite polymorphic (RAMP) DNA and microsatellite (MS) DNA to analysis of the hereditary structure and association. Main research results as follows:1. This study applied to the measurement to mtDNA D-loop area segmental sequence at 12 Roe deer. A 614bp sequence fragment from 12 individuals was obtained. There were 40 poly morphic sites in comparable 562bp sequence fragment, the rate of polymorphic sites was 7.12%. Among them 20 were singleton variable sites, 20 were parsimony informative sites, 38 were transition, and 2 were transversion. Besides, there were 2 undetected sites, 3 sites with alignment gaps or missing. Altogether 12 haplotypes were detected, sequence differentia averaged at 2.00% between haplotype. Nucleotide diversity (Ï€) is only 0.0204. Analysis of molecular variance (AMOVA) shows, having 58.09% variation come from the same population, 20.58% variation comes from individual among area department, 21.33% variation comes from population within area department. The northeast Roe deer of three areas department approximately classed three populations via the calculation of hereditary distance and cluster analysis.2. In RAMP molecular marker analysis, 16 couple primers from 112 random ones were used to amplify 31 Roe deer's samples. Obvious bands from three to nine were observed from each amplified primer, the average is 5.4 bands. Eighty-three bands bringed polymorphism among 87 bands tested and polymorphic percentage was 95.4%. Six bands were accordant from 3 couple amplified primers, none polymorphic. Others have abundant polymorphism. The UPGMA cluster analysis revealed that the result accorded with mtDNA D-loop area examined, for example, populations of Chang-bai-shan and Da-xing-an-ling attribute to own respective populations, there...
