L. spicata (Thunb.) Lour. var. prolifera Y. T. Ma is one main original plants of the traditional Chinese materia medica "Shanmaidong". Its tuberous roots are used medicinally, with effects of nourishing Yin to promote the production of body fluid and moistening the lung to clear away heart-fire. L. spicata var. prolifera is one geoherb from Hubei province. In this paper, In order to develop and utilize the resource of L. spicata var. prolifera, the resource-quality and the genetic diversity were studied.It provides available data and theoretical foundation for breeding. Our findings have important practical significance for increasing production and quality and raising industry of L. spicata var. prolifera. The results were as following.1.Quality evaluation of different Seedling from L. spicata var. proliferaTo provide the scientific basis for standard operating procedure,we study the effect of different seedling on the growth and active constituents of L. spicata var. prolifera.There were significant differences between the no.3 seedling and the no.1 seedling.their fresh weigh is 1151.30 kg per acre and 862.92 per acre respectively, but 2,4 seedlings were no significant differences, the correlation coefficient is 0.714 and 0.645 between Seedlings、dry weight and fresh weight, It show that the different seedling directly affects production, and the influence of dry weight than that of fresh weight. There were no significant differences between the Content of polysaccharide and L.sides B, they are at 20% and 2.00-2.93×10-2% respectively.The same as between soil and maidong of paclobutrazol residue. Taken together,the fine seedling:seedling with coarse roots and newborn buds, seedling with coarse roots, rootlets and newborn buds2.The influence of soil factors on L. spicata var. prolifera.The growth status of L. spicata var. prolifera. is closely related to environmental conditions.And the quality of Maidong is determined by many factors including microbe and soil.the content of pesticide residue and heavy metal was lower under GAP,The results showed that continuous cropping had no significant effect compared with the virgin soil except yield,the relationship among the dry weight and fresh weight of maidong and Soil physical and chemical properties has strong correlation, especially organic, pH value and available potassium. correlation coefficient is0.751、0.817and 0.616.in, it was significantly that correlation coefficien between Polysaccharide、effective phosphorus and pH value c is 0.419 and 0.516 in effective component aspects,The content of L.sides B and effective phosphorus and total potassium is 0.648 and 0.435. it have nothing to do withthe most physical and chemical properties.because of continuous cropping, the quantity and variety of Soil microbial has changed, it presents that bacterial soil translate into fungi soil.3.Somatic embryogenesis and plantlet regeneration of L. spicata var. prolifera.Somatic embryogenesis and regeneration plantlets were successfully induced from leaves of L. spicata var. prolifera. by tissue culture.The effects of Various factors to growth speed of calli and somatic embryogenesis were investigated. Up to 84.3% of explants produced embryogenic callus and embryogenic structures on MS medium supplemented with BA 2.0mg·L-1+NAA 0.5 mg·L-1+AgNO31.5 mg·L-1 the regeneration plantlets and somatic embryogenesis were rooted on MS medium with BA 2.0 mg·L-1+NAA 0.3 mg·L-1. The plantlets with root were successfully transplanted to the medium with vermiculite and ruby mica and humous and the survival rate could be up to 94.12%. Histological observation proved that the somatic embryo structure formation.4.Studies on the genetic diversity of Liriope spicata Lour. var. proliferaThe genetic diversity and genetic structure of L. spicata var. prolifera were analyzed by RAPD (Random amplified polymorphic DNA) and ISSR (inter-simple sequence repeat).primers were screened and a total of 215 DNA fragments were amplified, among which 116 were polymorphic, i.e. the percentage of polymorphic bands (PPB) was 53.95%.The observed number of alleles (Na) and effective number of alleles (Ne) were 1.5364 and 1.3891, respectively. The average values of Nei’s genetic diversity index (He) and Shannon polymorphism information index were0.2179 and 0.3174.The total genetic diversity (Ht) was 0.1910, and the population gene diversity (Hs) was 0.0815. The coefficient of gene differentiation among populations within species (Gst) was 0.5731, suggesting that intra-population variation is the main source of the genetic diversity of L. spicata var. prolifera.the cluster analysis showed that the genetic sibship among populations was correlated with geographical location. The zhognxiang and yicheng populations showed the highest diversity, while the oumiao population had the lowest diversity.5.The systematic position of L. spicata var. proliferaThe study chose Lriope and Ophiopogon to analyze genetic diversity and sibship by ISSR, in order to compare genetic differences within a genus. the genetic coefficient of L. spicata var. prolifera and L. spicata in local is about 0.7, relative to L. spicata in other areas is about 0.5.the relative relationship of L. spicata var. prolifera, between L. spicata and wild Ophiopogon japonicus was more near.It is possible that L. spicata var. prolifera, as triploid, could hybridize naturally between Lriope and Ophiopogon or with in Liriope.
