The Study Of Inducing Lily Polyploid By Several Chemical Reagent

The polyploid lily seeding which have doubled chromosomes are always strong high in quality and have fat scales. The study of doubling the chromosomes of lily not only has great academic meaning but also ecomomic value.In this study , We use colchicines and some pepticides to induce the chromosome-doubling of Novena. At the same time, we also recearch why Fangio pollen is yeld and how to double its chromosome; observe the process of bulblet growing from cutted-scale.Finally, we conduded that:1. Triflurclin has little effect on Novona dubling after the trifluraline-treatment, a few seedling differentiated; they have freshly leaves, and more than 24 chromosomes, some of them are teraploid. The scales treated by 0.005% trifluraline for 24 hours have high variation rate. But most tube-seeding browned or died, only a few variated. The effected treatment is trifluraline 24hours and 36hours.2. Take Novona scales as test pieces, after treated by colchicines, some of the seedling browned or died, also a few seedlings variated. They appeared fleshy scales when fostered in rootage medium; we use the root tip tabletting and found some have more than 24 chromosomes, a few of which are teruploid. 0.1% colchicines for 24hours; Siberia ,0.05%,72hours;Fagio,0.1%,72hours or 0.3%, 48hours( but also get high death rate).3. Take Novona tub-seedling as test pieces, after treated by colchicines, the same phenomenon appeared as the tested Novona scales. And Novona got high variation rate treated by 0.1% colchicines for 48hours; Siberia: 0.1%,24hours(But most of them browned or died);Fagio:0.1%,48hours got high variation rate.4. Triploid Fargo's karyotype is 3B, it has 36 chromosomes which are R(2n)=3x=3m+9m(SAT)+3sm+3sm(SAT)+9st+3st(SAT)+3t. The abnormal meisosis such as appearance of chromosome bridge, out of step, unequalization are the main reasons why the pollen of Fagio is viabilitiless.5. By parattin section, we became conscious of that first cell groups grow from the base of the cutted scales, cell groups grow into bud. Both sides of the bud primodium grow into leaf primodium. Finally bublet grow into leaf primodium.
