Study On The Inhibitory And Promotional Effect Of Exogenous GA3、ABA Upon The Dormancy Of Lanzhou Lily Bulb And The Physiological Mechanism

This experiment took Lanzhou lily as the test materials. We disposed the ?Lanzhoubaihe No.1?which was stored on the low temperature(2℃) condition by the Exogenous Gibberellin(GA3) and abscisic acid(ABA) in order to study the inhibitory and promotional effect of the germination of Lanzhou Lily bulb. Through the analysis of the content of carbohydrate, soluble protein, endogenous hormones(GA3, ABA, IAA, ZR),the changes of physiological,the biochemical indexes of antioxidant enzyme activity and phenolics synthesis related enzymes in bulb in the process of dormancy to explore the physiological mechanism of exogenous GA3 and ABA promoting and inhibiting effect on the sprouting of seedball. The results of the research could provide the basis for enriching lily bulbs dormancy mechanism of the core theory and Lanzhou Lily seed treatment technology. The main research of this experiment was as follows:(1)Under the condition of 2℃, GA3 treatment could shorten the germination time of Lanzhou Lily bulbs. The secondary ball germination time reduced from 105 days to 90 days. ABA treatment could inhibit the germination time of Lanzhou ball, and the third ball could prolong germination time from 95 days to 115 days. GA3 promoted the germination through promoting the growth of a ball terminal bud to enter logarithmic growth period early, and to increase the growth rate of the period and promote seedball germination. Therefore, ABA inhibited bulbs germination by delaying the growth of a ball terminal bud in logarithmic growth phase and inhibiting in the growth rate of the period of terminal bud.(2)In the embryonic period, starch content in bulb decreased with the lengthening of storage time under low temperature, and the content of soluble sugar, amylase activity and soluble protein content significantly increased with the storage time under low temperature; exogenous GA3 treatment accelerated material changes which was in the bulb above, while exogenous ABA treatment delayed the changes of material in the bulb. The length of terminal bud, the distance of bud, and the height of bud or bulb and soluble sugar content, starch content, amylase activity and soluble protein content were of correlation. The height of bud / bulb dielectrics in 0.5-0.6 can be a standard to judge the fact whether the Lanzhou lily bulb has been fully discharged or not.(3)During the process of Lanzhou Lily releasing dormancy, the activity of POD and CAT with different treatment in bulb decreased with the cold storage time, which accumulation of CAT and POD adjustment bulb and its related substances of lily, and it had the promotional effect on breaking dormancy. The activity of PAL decreased with the storage time of low temperature, and the activity of PPO was increased, which showed that phenolic compounds had inhibitory effect on breaking dormancy of Lily bulb. Treating with exogenous GA3 promoted changes in lily bulbs of the above 4 kinds of enzyme activity, while exogenous ABA treatment delayed the change of enzyme activity of Lily bulb.(4) In the process of breaking dormancy, the content of endogenous GA3, IAA, ZR increased rapidly, while the content of endogenous ABA decreased, at the same time, the ratio of hormones(GA3/ABA, IAA/ABA and ZR/ABA) with low temperature storage time increased. The secondary seedball treated with GA3 in low temperature storage of 20 days was the key point of the 4 kinds of endogenous hormone content and the hormone ratio change violently, that promoted these changes of endogenous hormone content. The third seedball treated with ABA during storage at low temperature 30-50 days was the key period of 4 kinds of endogenous hormone content and the hormone ratio changes, that restrained these changes of endogenous hormone content.(5) The correlation study of substance metabolism in the bulbs during dormancy release of lily bulb showed GA3 and ABA and the content of soluble sugar, starch, PAL and other indicators are relevant, so to further clarify the GA3 and ABA was the main hormone in breaking dormancy of Lanzhou Lily. At the same time, studying of bud length, bud distance, bud high / high bulb and endogenous hormones of each ratio correlation showed that GA3/ABA with bud high / bulb height was highly significant correlation. So GA3/ABA is 1.2-1.4 could be the standard of judging Lanzhou lily bulb dormancy has been fully discharged.
