| Cyclamen is a plant which belongs to Primulaceae, the genus Cyclamen. Cyclamen is widely cultivated as an important ornamental houseplant, having decorative leaves and showy, variously colored flowers with reflexed petals. The genus Cyclamen contains about 21 species. The present horticultural cultivars derived from cyclamen persicum Mill.Cyclamen is originated in south Europe and Mediterranean regions, which are oceanity monsoon climate. The resistance of cyclamen is lower,so its economic value has seriously been influenced under environmental stress. Many studies had examined that enhancement of Mn-SOD expression in transgenic plants could improve the stress tolerance of plants. In the study, we had introduced the Mn-SOD gene into cyclamen to evaluate its effect on the endurance of environmental stress. The transgenic plants derived from leaf of cyclamen explants would pave the way for its breeding via genetic engineering.The main results were as follows.1. The efficient adventitious bud regeneration system was set up for"scarlet 1011"cyclamen cultivar.The basal medium, TDZ and NAA were used to compare their effect on adventitious buds induction from leaves of"scarlet 1011"cyclamen cultivar by using the orthogonal experiment design. The results showed that TDZ played a determinative role in inducing the adventitious buds, the secondly factor is the basal medium and NAA. The ideal combination for leave regeneration medium was 1/3 MS+TDZ 0.5 mg/L+NAA 0.3 mg/L, on this medium, the leaves regeneration frequency was 93.1%,and the mean regeneration number of buds was15.7 . leaves age and leaves part have different effect on bud regeneration,using the leaves of 40d leaves age can enhanced the regeneration frequency of cyclamen, the regeneration capacities of leaves base part was more higher than others. No effect on regeneration efficiency of leave was found by adding up AgNO3 while its concentration varied from 0.1-0.6 mg/L, however, AgNO3 would reduced regeneration efficiency while its concentration was over 0.6 mg/L.Adventitious buds regeneration from hypocotyls, cotyledon, and petiole of aseptic seeding which come from"scarlet 1011"seeds were studied.
