Tomato is one of our main vegetables, and tomato seed purity and authenticity are very important for the better development of the tomato industry chain. To meet the standards of the international market, and to protect the legitimate rights and interests of farmers, we must develop an accurate, rapid identification method of hybrid purity. The main influence factor of hybrid seed purity identification is the false hybrids from female parent self-cross. In the base of researching'Jinpeng No.1'tomato RAPD marker, we convert it to SCAR marker for testing tomato hybrids purity, then determine single dry seeds as the source of template DNA, successfully established the molecular identification system that identified'Jinpeng No.1'tomato hybrid purity by SCAR marker.Compared 4 methods of genome DNA extraction, and used it respectively to RAPD and SCAR amplification, then found the method of Gong Zhen-hui (with minor modification) was better than the others, and settled the brown colored question of tomato DNA. Explored and improved the extraction method from dry tomato seed, and used it to the work of identification'Jinpeng No.1'tomato hybrid purity of SCAR marker.Explored various factors of RAPD reaction system; Optimized and established RAPD reaction system; cloned and sequenced the unique segments S130771 and S296943; successfully converted two unique segments to SCAR marker and designed two pairs of SCAR primers; the putative amplified DNA fragments were obtained from male parent and F1 hybrid by primer pair RF2/FF2, which is the same as the isolation of RAPD marker S130771; ultimately SCAR marker S130771 can use to identify the hybrid purity of'Jinpeng No.1'tomatoes .Established the purity identification system of Jinpeng No.1 tomato dry seed by SCAR marker, and detected 100 seeds, 85 seeds were successfully amplified.
