Five parents of hybrid japonica rice were put to the test including two male sterile lines named Hanfeng A,8204A , two maintainer lines named Hanfeng B,8204B and one restorer line R161. All these materials are backbone parent strains of hybrid rice popularized and widely applied in shanghai area.Five parents of hybrid japonica rice including Hanfeng A,Hanfeng B,8204A,8204B,R161 were tested for RAPD analysis with thirty-six random primers. Different RAPD markers among these cultivars were cloned and sequenced. Based on these sequence, SCAR markers were designed, synthesized, and confirmed in the rice cultivars. Totally eighteen RAPD markers were successfully conversed into six SCAR markers. The five rice cultivars and their derived hybrid F1 seeds with these SCAR markers were detected combined with a pair of cytoplasmic male sterility specific primers to distinguish two pair of sterile lines and maintainer lines. The results of seed purity from molecular detection were also confirmed by the field tests. These indicated that the developed molecular markers can be use to identify five hybrid japonica rice lines in laboratory.These results proved it was success that RAPD markers be converted to SCAR markers. The SCAR markers will be used to authentication of hybrid purity of japonica rice seeds from yields. Base of the DNA markers and a method using to extract DNAs from plants, a system in which hybrid purity of japonica rice seeds could be identified quickly, well and truly was build.
