Peanut is an important oil and industrial crop in our country, and plays a decisive pole in agricultural production and national economy. Increase of production, improvement of quality and economic performance are core job in peanut production and research in our country. Genetic improvement for peanut with the high and new technology is linchpin of getting breakthrouth in peanut breeding and reasearch in futrue. Growing up of DNA molecular marker breathe new life into the peanut reasearch. The use of the high and new technology in peanut is still little comparing with other crops. The molecular markers used in peanut berfore are RAPD and RFLP, the development of DAF, SSR, AFLP molecular marker provides more methods for DNA polymorphism reasearch of peanut. Such progress has been maked in linkage map construction of peanut at the same time, and some linkage maps had been constructed, but these linkage maps were constructed base on wild species mainly, one linkage map which based on cultivated species in the whole meaning is not constructed until now. Great difficulty exists in reasearch of some important economical character and economic character of peanut cultivated species with these linkage maps. Therefore it is necessary of the construction of population based on peanut cultivated species for construction of linkage map. Linkage map for tetraploid peanut(cultivated species)were constructed by the new SRAP molecular marker in this study. The results were as follows: 1. The application of SRAP in peanut molecular analysisSRAP (Sequence-related Amplified Polymorphism) is a new marker system based on PCR and preferely amplifies ORFs (open reading frames) by special primer design. SRAP combines simplicity, reliability, moderate through-put ratio and facile sequencing of selected bands and have been applied in map construction, comparative genetics, genetic diversity and gene mapping.In this study, SRAP was applied in peanut genome analysis. We refered to the PCR reaction system which had been stable in cotton and mixed BSA into the system in the primer screening research, and then the system was tested and improved more and more. At last we developed and optimized an efficient PCR reaction system for detecting SRAP that showed reliable, repetitive and productive. 2.
