Genetic Linkage Map Of Anthurium Andraeanum Based On SRAP Molecular Markers

The molecular linkage map of Anthurium andraeanum was constructed using the method of‘double-pscudo-testcross’ with the SRAP data obtained from the F1population of cross combination‘Pink Champion’בDakota’. The ‘Pink Champion’ was very resistant to stresses, and the ‘Dakota’ hasgood integrated traits, such as plant appearance and color. The construction of the map will provide abase for the QTL location, gene clone and MAS research for Anthurium andraeanum. The main resultswere summarized as follows:1. Construction of F1populationThrough the way of artificial pollination, seed disinfection, in viro culture and the seedlingsacclimatized under greenhouse condition, a F1population of152individuals from a cross betwecn‘PinkChampion’ and ‘Dakota’ was obtained, of which94individuals were selected to construct the map.2. Optimization of SRAP reaction systemBy using ‘Pink Champion’ and ‘Dakota’ as the template DNA and orthogonal experimental design,the factors which affected the SRAP amplification of Anthurium andraeanum were studied. The resoultsshowed that the optimal reaction system was: the20μL reaction system contained40ng template DNA,0.8mmol/L upstream primer and downstream primer,2.5mmol/L Mg2+,0.2mmol/L dNTPs,0.5U TaqDNA polymerase.3. The SRAP analysis of F1population209pairs of SRAP primers were firstly selected from the parents, and then according to thesegregation of hybrids,141primers which were clarity, stability, and of good amplification wereabtained to construct the linkage map. They amplified a total of355polymorphism loci, and each pairof primers amplified2.4loci on average. The polymorphism loci were mainly between50bp and500bp.4. Construction of the genetic linkage map of ‘Pink Champion’בDakota’The genetic linkage map of ‘Pink Champion’בDakota’ was obtained by utilizing JoinMap4.0software’s CP model. It included19linkage groups which contained254loci and covered genome1689.5cM with a mean marker interval of6.65cM.
