| During the experiment using two cell lines of Pinus strobus (No.01 and No.02) to induce mature somatic embryos, it was found that only the No. 01 cell line had the capacity to form mature embryos. So, we attempted to predict earlier ET's potential ability to form the mature somatic embryos through comparing differences in ET's morphology and structure during proliferation culture. By analyzing ET growth, sucrose metabolism and protein expression during the proliferation stage, we aimed at probing into the morphology, cellular and molecular basis in the potential of inducing mature embryos. The results were as the followings:(1)The two cell lines had no significant morphological differences in proliferation stages, while had difference in the structure: No.1 cell line had a type of structure named ESM4 which was lack of in No.2 cell line.(2) From the RGR curves of ETs, it was found that the RGR of cell line No.2 was higher than No.1cell line throughout the whole proliferation culture stage and reached to maximum during the forth and fifth day, which had a delay compared to No.1 cell line. The RGR of No.1 cell line reached to maximum on the third day and it was very low on the 15th day and was closed to 0 on the 20th day.(3)There was some correlation among hexose contents, the rate of hexose/sucrose, activities of Inv enzymes and RGR.(4)The total soluble protein contents were measured by Bradford's method. Generally speaking, No.1 cell line was significantly higher than No.02 in total soluble protein content. |
